Taxonomy Of Two Kinds Of Freshwater Cnidarians In Hebei Province And One Kind Of Freshwater Cnidairans In Hunan Province

In the present study, three kinds of Cnidarias were collected from Hebei and Hubei.They were analysed by DNA barcoding methods combined with morphological features. Themain results as follows:This paper reported a new record of C. sowerbyi in Hebei Province.18S rRNA andrDNA ITS genes of the Craspedacusta sp. were amplified and sequenced．The geneticdistances were calculated and phylogenetic trees were constructed using the software MEGA5.0. The results showed that the rDNA ITS of Craspedacusta sp. from the Hebei were highlysimilar to those of C. sowerbyi (The maximum similarity100%)．The minimum geneticdistance was0. And Craspedacusta sp. was belong to the C. sowerbyi in the phylogenetictrees．Except the minimum genetic distance was0.029, the remaining results based on theanalysis of18S rRNA were in accord with the above. The morphological features of jellyfishwere described.This paper reported a new record of H. vulgaris in Hunan Province. mtDNA COI andrDNA ITS genes of the Hydra sp. were amplified and sequenced．The genetic distances werecalculated and phylogenetic trees were constructed using the software MEGA5.0. Based onthe analysis of rDNA ITS, The genetic minimum distance was0.003. And based on theanalysis of mtDNA COI, The genetic minimum distance was0.005. The morphologicalfeatures of Hydra were described.The C. sowerbyi collected from the Hunan University, was a new distribution in Hunan.Both mtDNA COI and rDNA ITS genes of the Craspedacusta sp. were amplified andsequenced. Based on the analysis of rDNA ITS of the Craspedacusta sp. form Hunan andHebei, The genetic minimum distance was0.016. And based on the analysis of mtDNA COIof the Craspedacusta sp. form Hunan and Hebei, The similarity was97%.19polymorphiclocis were all the transitions.
