With rapid development of synthetic biology, in the field of engineering applica-tion, huge demand for designing exogenous gene which will be transfered into car-rier cell to make it express in host cell to guide some metabolic pathway’s synthetic in order to get target products.This is already an important method to get product that can’t be got from nature in quantity. The design of metabolic pathway synthesis is a hot aspect that its development may play a vital role in more and more filds like energy filed and so on.Although many researchers have complete the work of metabolic path synthesis in the experiment,there is no tool for assisting related works especially.The work con-sists of pathway design,synthetic gene design,oligonucletode design an so on,each parts has its own focus based on different exprimental environment and purpuse while some problems still can’t be explain clearly on each part. In order to design highly expressing DNA suquence to guide metabloc pathway’realizing in host cell,we pay attention on every aspect of this work.In this article,we mainly do three work:1.try to find the relationship between the feathers and the fethers with expression levels,here we analyze the feathers of NDA seuqence that may affect the expression level to realize the work;2. develop a new method of oligonucletode design for Gene Chips and a mtheod for reversing AA se-quence to DNA sequence,with learning olgonucletode design algrihtms for PCR or LCR and traditional protein-to-DNA algtithms on different demand;3. integrate the whole system for metabolic pathway systhesis’design,it can be used to complete the whole work. At last,we use an example to explain the result of each part and the rsults can prove that the sequence deisgned can satisfy the setted parameters,the systerm provide a good result.
