Due to the importance of the long-chain n-3polyunsaturated fatty acids (n-3PUFAs) on human health, many studies have been published in the productions of fat-1transgenic mice and domestic animals. In this study, we used a different fatty acid desaturase gene, fad3b derived from flax (Linum usitatissimum) to create transgenic mice. At first, the fad3b was connected to the mammary gland specific expression vector, pBC-asl, then the vector was digested with M. Mun I and Apa I, and a marker-free and resistant gene-free and mammary gland specific expressed fad3b transgenic box, pBC-asl-fad3b was obtained. The length of pBC-asl-fad3b expression box is12188bp, which avoid the marker genes, resistant genes and bacterial frame sequence and ensure the transgenic biosafety. Using this transgenic box the transgenic mice were created by pronuclear injection and the mice have produced3generations. Analyses of the transgenic mice showed that the expressions of fad3b significantly expressed in the founder, F1, F2and F3generation mice. We also examined the expressions of4genes associated with fatty acid synthesis and metabolism, FASN (fatty acid synthase), HSL (hormone-sensitive lipase), LPL (lipoprotein lipase) and ACC (acety-CoA carboxylase), in various tissues of the transgenic mice.
