The large quantity and volume as well as the appearence in many geological ages make the fluid inclusions in halite as the important material to study the ancient atmosphere, hydrosphere and biosphere. It can provide reliable quantitative data to the research of paleotemperature, ancient sea water and atmosphere composition, and the creature evolution. Recently, the foreign researchers have done a lot of work in the analysis method of fluid inclusions in evaporite formed in the surface condition. But few can see in China. So we have done some exploratory study in the homogenization temperature, petrography, the mechanism of entrapment halophiles, and the extraction the DNA of halophiles of the fluid inclusion. We got large amount of experimental data and robust results. Here is the result:(1) We use cooling nucleation method to measure the homogenization temperature of the fluid inclusion in halite evaporite at50℃. The max homogenization temperature coincide with50℃. That means the max homogenization temperature can reflect the paleotemperature which the fluid inclusion forms. The homogenization temperature and the size of of the primary fluid inclusion shows no correlativity.(2) By using the cooling nucleation method, we get the paleotemperature of Northern Shaanxi Salt Basin in the O2m65stage of Ordovician period is about27-36.9℃. The paleotemperature is higher than the modern temperature of this place. The one reason may be the plate which Northern Shaanxi Salt Basin located in has moved from the low latitude in Ordovician to the middle latitude. Th other one is the paleotemperature of mid-Ordovician is higher than the modern temperature according to the isotope geothermometry. The result is consistent with the Oxygen isotope geothermometry.(3) The size of fluid inclusions with halophiles is bigger than the one without them. If the halite crystal has more fluid inclusions with halophiles, the more halophiles in each fluid inclusion will be. The temperature of evaporation may impact the density of halophiles in a fluid inclusion. But it is not relevant for the shape of the halophiles. So does the origin density of halophiles in the brine.(4) We build the method of molecular biological technology in fluid inclusion entrapment. We successfully extract the DNA of halophiles in fluid inclusion and demonstrate the origin of DNA by gene sequence BLAST.
