Objective:In a preliminary experiment,we found out the interacts of RTN3with FBLN5by carrying out yeast two-hybrid screen, using RTN3as the bait to find its potential associated candidates.By the gene level, protein level and cell level, we try to figure out whether RTN3FBLN5interaction prediction and verification. Related research is to explore RTN3apoptotic protein functionality in addition to other features provide clues.Methods:(1)Through bioinformatics methods, by analysising RTN3gene sequence, we predict RTN3gene promoter sequences and transcription factors;(2) By structure modeling of RTN3and FBLN5protein,to predict their interactions possibility;(3)From the cellular level,using of co-immunoprecipitation techniques,we confirmed wether RTN3can interact with FBLN5;(4) Based on bioinformatics analysis,partial deletions constructed RTN3carrier protein for functional RTN3research conditions. Results:(1)By gene sequence analysis,we forecast RTN3promoter sequence located near the47-187bp,and transcription factor is GATA-1;(2)Through protein structure prediction,we confirmed RTN3has the structure of interaction with FBLN5;(3) Through co-immunoprecipitation experiments,we confirmed that the interaction of endogenous RTN3with FBLN5can occur.Conclusions:(1)Through bioinformatics analysis the structure and function of RTN3and FBLN5,we can predict the interactions occur;(2) We confirmed the RTN3in smooth muscle cells can interact with FBLN5.
