Apios americana Medikus is a kind of leguminous plants. It has been proven that theApios’s tubers is effective in inhibiting cancer and lower blood pressure. But it is rarely to seeany research on Apios americana Medikus in domestic.This article tries to use the Apiosamericana Medikus to product ACE Inhibitory Peptides by enzyme hydrolysis method.The maincontent of this paper includes the following aspects:1.Find out the optimum process for protein hydrolysate. The result was that:the optimumcondition was:62℃, pH8.5, substrate concentration was1.2%, enzyme dosage3000u/g.Andthe result of amino acid composition analysis shows that the enzyme hydrolysation do notchange the amino acid composition of the sample.We use SepHadex G-25to find out themolacular weight of the digest product. The result shows the alkaline protease have good effecton digest the Apios protein.2.Ultrafitration and desalination.The result shows that the Peptidepe whose molecularweight below3kda show the high activity in inhibitor ACE. And the mass fraction of thesolution for ultrafitration is0.8%and the pressure of ultrafitration is250kpa. Duringdesalination the elution fractions of70%ethanol solution do better in inhibitor ACE activity theIC50is0.187mg/mL.3.Prificate the product by DEAE.Sepharose Fast Flow and Sephadex G-15.AfterDEAE-Sepharose Fast Flow there are three elution peaks. The third peak is most activities. It isfurther purificated by Sephadex G.15.During the tow peaks,the second shows high activity.Itsfinal IC50is37.47μg/mL.4.The chemical properties detection on the peptide shows it has good foamability but itsfoam stability is low. The emulsification of the product is100%,8hours later,the emulsionstability is54.2%. Besides, it is stability on heat and photo treatment. The β-cyclic dextrin caninfluence the peptide’s activities while the other kinds of food additives has rarely influence onit.The in vitro digestion experimental prove that the products has good digestion stability.
