This report explains the working principle of optical tweezers and the optical trap force calculation method of Mie particles and Rayleigh particles.At the same time, it sums up the calibration method of optical trap stiffness at the same time. On the premise of the optical tweezers capture particles stably, I built an optical tweezers system which can be used to manipulate cells by moving2D stage and light beam according to the basic components of the optical tweezers system.The escape velocity is an important parameter to predict and interpret the physical properties of cells based on the understanding of the cell test applications of optical tweezers. An optical tweezers system is designed to capture and test cells. In the experiment, it first ensures the cell is stably captured by the control of the light beam and the stage. And then I test the escape velocity of cells. The sample cells are kept in constant motion with two-dimensional stage in a culture environment to improve the fluid force method for the calibration of optical trap stiffness and the escape velocity is known.The report is achieved that the cells are trapped by optical tweezers stably in liquid. And the escape velocity is tested. The fluorescent calibration is not needed for the cells to keep cells alive and implement the test of living cells.Therefore, the optical tweezers can effectively solve the drawbacks of the medicine in the cell test.
