Long chain fatty acids have a wide range of physiological functions in mammals, especially playing an indispensable role in biofilm formation and maintenance of dynamic characteristics. Fatty acid transport protein1(FATP1) and plasma membrane fatty acid-binding protein (FABPpm) are plasma membrane proteins and expressed in many tissues and organs with active fatty acid metabolism, e.g. heart, liver, muscle, and small intestine. It has been shown that FATP1and FABPpm play an important role in the uptake and metabolism regulation of long chain fatty acid, but there are fewer reports on mechanism in the mammary gland of ruminants.In this study, the full-length cDNA sequences of FATP1and FABPpm gene in the mammary gland of Chinese Holstein cattle were cloned by RT-PCR and RACE technology. Then the cDNA sequence and the amino acid sequence and protein structure of FATP1and FABPpm were analysed by bioinformatics. The results showed that the full-length cDNA sequence of FATP1has3459bp, which contained a1941bp of open reading frame(ORF) encoding646amino acid residues, flanked by a243bp of5’-Untranlated Regions(5’-UTR) and a1275bp of3’-UTR; The full-length cDNA sequence of FABPpm has1700bp and its ORF has1293bp, encoding430amino acid residues, flanked by a108bp of5’-UTR and a299bp of3’-UTR; The FATP1gene encode a protein with a relative molecular weight of71.0238kD and theoretical isoelectric point of8.83; The relative molecular weight of FABPpm is47.5135kD and theoretical isoelectric point is9.19; The Secondary and tertiary structure of FATP1and FABPpm are mainly composed of alpha helix, extended strand and random coil; In comparison with the amino acid sequences of known mammalian FATP1and FABPpm, the amino acid sequences deduced from the cDNA sequences of FATP1and FABPpm of Chinese Holstein cattle showed different levels of homology ranging from88%to94%and93%to96%, respectively; Sequence alignment and phylogenetic tree analysis showed that the relationship of FATP1and FABPpm between Bos taurus and Sus scrofa was closest; Cloning the full-length cDNA sequences of FATPl and FABPpm gene in mammary gland of the Chinese Holstein cattle provided the basis for further study of their subcellular localization and transport function of long chain fatty acids.
