Grps have been found in the cell walls of many higher plants and form a third group of structure protein components of the wall in addition to extensins and proline-rich proteins. The main biological nature is that strengthen the stability of cell wall structure and helps the formation of drawn fibers. ZmGrp3is a root-specific gene which isolates and characters from maize.Plant roots are importance of nutritions and water absorption, storage, transportation. So, the study of ZmGrp3gene and promoter has wide application value.The study mainly includes he construction of expression vector expression and analysis of function in the ZmGrp3gene promoter and the analysis of the ZmGrp3gene’s structure and polymorphism. The results are summarized as follows:1、ZmGrp3genes were cloned by PCR from different maize inbred line.Using the bioinformatics to analysis the structure and the nature of protein, We found that the protein is hydrophobic protein which contains glycine, alanine, cysteine, and threonine, and glycine accounted for40-50%of the total protein. The secondary structure of the protein is alpha-helix, beta-folded and random coil.2、The analysis of sequence from21inbreds,we can know that the divergence between genes is small which caused by the role of purifying selection, and a small number of positive effect.3、A2241bp fragment of the5’upstream region of ZmGrp3gene promoter was obtained by PCR from the maize inbred line B73(Zea mays). The result of promoter sequence analysis indicates that it contains TATA-BOX, CAAT-BOX, GATA-BOX Cis-acting elements and AGCT core sequence.4、The new plant expression vectors were constructed by placing the CaMV35S promoter of pCAMBIA1301with GUS reporter gene. Then the plasmids containing the fusion constructs were transferred into rice zhonghua11by Agrobacterium-mediated transformation. Through screening and molecule detection, harvest transgenic plant.5、Through Gus dyeing, the ZmGrp3gene promoter can express in the root tip stem and leaf of rice.
