Environmental microbes, whose geochemistry function mainly displays in the rock and mineral weathering, element migration and accumulation, organic matter degradation and deposit formation, etc, participate in organic and inorganic matter circulation and affect their global distribution, thus playing an important controlling role on substance in the upper lithosphere, hydrosphere and the distribution of atmosphere, on the earth since its formation. In recent years, along with establishing and developing the nucleic acid extraction and analysis, researchers began to apply molecular biology techniques for the analysis of environmental microorganisms, such, partially overcomes problem of those traditional technique, by which not only cultured strains were difficult to be clearly classified and uncultured bacteria cannot be cultured and studied.With the use of molecular biology techniques, the unpredictable, complicated and long process in strain isolation, culture, phenotypic screening is no long unavoidable, and through directly DNA or RNA sample analysis, microbial genetic information and other information accessable were maximized, thus a comprehensive and rapid analysis of environmental microbial diversity is affordable. While, the use of molecular tools to study microbiathere takes an premise on, obtaining DNA or RNA with quality over baseline.Involvingly, research in this paper intends to optimize an simultanous acidophilic bacterial DNA and RNA extraction protocol, and measuring extraction effect, to preliminarily explore the best extraction conditions of several levels in some categories. At first, while Acidithiobacillus ferrooxidans (referred to as A.f) as the research object, LiCl and TRIzol method tested, several degradation of DNA reavealed in LiCl method tests and TRIzol method was chosen then.Acidithiobacillus ferrooxidans material expertiments kept on, the influence of extration buffer with different pH, different concentration protease K, biomass amount on extraction effects was revealed. The one with pH7.0buffer and without protease K performed best in their group. And the amount of nucleic acid increased after biomass increase allover tested range.As following, additionaly, extraction effects on acidophilic microbe, Thiobacillus thiooxidans, Thiobacillus caldns, Leptospirillum ferriphilum, Ferroplasma thermophilum studied respectively, and universality of the protocol was partly explained resultedly.
