Abstract/Summary:
In this article, the mechanisms of the cerium oxide and yttrium oxide nanoparticles onthe proliferation of primary osteoblasts were studied. The physical and chemical properties ofnanomaterials were characterized by SEM, XRD and DLS. The proliferation of osteoblastswas measured by MTT method. Then the promotion mechanisms of cerium oxidenanoparticles on the proliferation of osteoblasts were investigated from cycle regulation, celladhesion and oxidative stress. The inhibition mechanisms of yttrium oxide nanoparticles oncell proliferation were researched through the change of intracellular reactive oxygen specieslevel, mitochondrial membrane potential and cell apoptosis rate of osteoblasts. The resultsshowed that:1. Cerium oxide nanoparricles with two sizes can promote the proliferation of theosteoblasts. Cerium oxide nanoparricles with30nm have a higher promotion effect than thatof60nm.2. Cerium oxide nanoparricles can stimulate more cells to pass through the G1/Scheckpoint into S phase, but did not change the length of cell cycle. Furthermore, ceriumoxide nanoparticles can also increase the adhesion of osteoblasts, thus promote cellproliferation.3. Cerium oxide nanoparricles can relieve the oxidation damage induced by hydrogenperoxide. The protective effects were increased with the decreasing size of nanoparticles.Cerium oxide nanoparticles protect osteoblasts against hydrogen peroxide induced oxidativedamage, suppression of antioxidant enzymes depletion and cell apoptosis via its reactiveoxidative species scavenging mechanism.4. Yttrium oxide nanoparricles can inhibit the proliferation of the osteoblasts and theinhibition effect was increased with the decreasing size of nanoparticles. The cytotoxicity ofyttrium oxide nanoparricles followed a size-dependent manner. Yttrium oxide nanoparriclescan increase the intracellular ROS levels, induce cell apoptosis, and then inhibit cellproliferation.
