Atomic Force Microscopy (AFM) as a surface information analysis tool has a highresolution and simple request to sample, becoming good research tool in the field ofmicro and nano processing and biomedical. Can realize the cell surface microstructureobservation, mechanical properties analysis and surface micro processing inphysiological conditions. Complete the exploration on the mechanical characteristics ofcell. This paper establishing the mechanical model and achieve change rule intheoretical analysis of cell internal structure and mechanical properties. At the sametime, experimental study of mechanical properties and micro-cutting is carried out.First, making a heater which can suitable for AFM, removing liquid beads whichproduced in AFM viewfinder, clamping generator and probe. Installing the heatingplatform and connecting the cooling circulation system to make liquid environmenttemperature keeping37℃. We get the normal cell and cancer cell surface morphology,adhesion force and elastic modulus. The results show that the elastic modulus of normalcell is larger and adhesion is smaller.Then, keeping the modulus and adhesion as known quantity when using theABAQUS finite element analysis software platform to establish three-dimension cellmodel and simulate the indentation force. The influence rule of modulus and adhesionto amplitude, maximum force and maximum adhesion can be got.Finally, using the gained interaction force to set scanning voltage and load voltage,then micro processing images of normal liver cells and hematoma cells and the relationcurves between load voltage and depth of processing can be obtained. The change ofcutting layer adhesion property can also be got. Results show that processing depth ofnormal liver cells deeper.
