Sphagnum is the dominant plant in peatlands, and it plays an important role in globalcarbon cycling. Different preservation conditions may affect spore germination conditions ofSphagnum which affects regeneration of bryophyte and vegetation pattern, even affectscarbon accumulation of peatland, The study of the spore germination conditions underdifferent preservation conditions is significant to maintain the ecological function of peatland.The current research in this field is rarely reported.This paper taking the spores of many kinds of Sphagnum collected from Hani Peatland inChangbaishan mountain as materials. Through different preservation conditions in field andlaboratory, this study researched the effects of different preservation conditions andenvironment on spore germination of Sphagnum. Taking the Sphagnum buried in differentlatitude peatland as the research object in this paper, study the effect of latitude and changesof water level on spore germination; Taking S. palustre, S. capillifolium and S. fallax capturedfrom2008to2011as the study species, to study the effect of preservation time on sporegermination of Sphagnum; Taking S. palustre, S. capillifolium and S. fuscum as theexperimental material, preservating in different temperature before culturing, addinggibberellin hormone in the prosess of culturing, study the effect of preservation temperatureand gibberellin hormone on spore germination of Sphagnum. Acquiring the followingconclusions through researching:(1)Changes of elevation gradient have an effect on spore vitality of S. capillifolium, Withthe decline of the altitude, spore germination rate of S. capillifolium has certain volatility;Water level conditions have an effect on spore vitality of S. fallax, spore germination rate of S.fallax in hollow is higher than hummock.(2) Changes of latitude gradient have an effect on spore vitality of S. palustre, S.capillifolium and S. fallax, With the decline of the latitude, spore germination rate of threesphagnum is on the rise. Water level have an effect on spore vitality of S. fallax, sporegermination rate of S. fallax in hollow is higher than hummock.Water level have an effecton spore vitality of Sphagnum which can be changed by latitude.(3)Different preservating time for different species, spore vitality is different. sporegermination rate of S. palustre, S. capillifolium and S. fuscum collected in2011is highest,With the preservating time extension, the germination rate shows a trend of decline; Sporegermination rate of S. capillifolium has certain volatility.(4) preservating temperature strongly affects spore germination of S. palustre, S.capillifolium and S. fuscum. With the reduction of preservation temperature,spore germinationrate of three sphagnum showed downward trend; Gibberellin inhibited spore germination of S.palustre, S. capillifolium and S. fuscum.
