Cell differentiation and polarity is the hot issues of life science research, to understand the individualdevelopment, disease and even cancer has important significance. At present, to control the molecularmechanism of cell differentiation and polarity changes, especially the key molecules, still know very little.Reelin stop signal, as a kind of cell migration, about the formation and development, layers of cerebralcortex nerve cells migration play a very important role. We use normal mice (WT) and reeler in micehippocampus slices-cell co-culture respectively, under the microenvironment of the Reelin protein in thehippocampus as signaling molecules mediates the degree of differentiation of cell differentiation andpolarity changes. This is we are going to delve into the hippocampal slices from molecular mechanismsmicroenvironment for cell polarity and differentiation.Objective: to observe the micro environment in hippocampal slices, reelin cells of differentdifferentiation degree of polarity and differentiation.Methods: respectively with the wild type (WT) and type reelin gene knockout (reeler) in micehippocampal slices with five different differentiation degree of the cell (liver cancer cells and normal livercells and PC12cells, and normal nerve cells and bone marrow mesenchymal stem cells) to jointly develop,observation of cell differentiation and polarity.Results:(1) low differentiated cells of the hippocampus slices microenvironment polarity anddifferentiation, can the influence of well-differentiated cells differentiation ability can no longer divide.(2)hippocampal slices of cells have the dual role of the microenvironment can promote cell differentiation, cellcontact inhibition with the slices at the same time, inhibit the growth of its own division and.(3) in the CA1area and dentate gyrus of signaling molecules can contribute to the increase of cells and brain slices ofaffinity, and cells can be induced by deformation and migration.(4) reelin has certain inhibitory effect oncells, prevent foreign cells to inhibit the growth of its polarity changes and at the same time, the status offoreign cells in neural cells induced differentiation and development, promotes the ordering.Conclusion: hippocampal microenvironment polarity and differentiation of cells, has inhibition to thegrowth and division, cell polarity changes related to themselves and micro environment, especially for low differentiated cell polarity and differentiation is larger, the influence of the reelin to cells, the same effect ofthe reelin may be factors that result in the change of cell polarity.
