Bacillus pumilus as a facultative Oligotrophic Bacteria, can be widely applied to detecting aquaculture water remediation and pollution. The viability and stability of probiotics are the two largest challenges producer in terms of technology and market facing. The spore has great resistance to various unfavorable environments. Through this experiment, optimized Bacillus pumilus vegetative cell culture medium and condition, the Bacillus pumilus spore culture technology research, improve the spore yields, All these lay a foundation for its application in production.According to the results of single factor optimization, orthogonal design has been used to optimize medium concentration and culture condition. The optimal medium: sucrose4%, peptone2%, NaCl0.5%, KH2PO40.1%, CaCl20.03%, MgSO4·7H2O0.01%, MnCl20.03%, FeSO4·7H2O0.004%, and the optimum culture conditions:fermentation temperature28℃, initial pH7.0, rotational speed180r/min, inoculation2%. After optimization, Bacillus pumilus biomass stability in220×108CFU/mL.Based on the results of single factor optimization, the concentration of sporulation salt solution and culture conditions were optimized by orthogonal design. The optimal medium:NaC10.5%, KH2PO40.25%, CaCl20.02%, MgSO4·7H2O0.02%, MnCl20.015%, FeSO4·7H2O0.004%, optimum culture condition:32℃, initial pH8.0, rotational speed180r/min, mixing ratio5:5. After optimization,the amount of Bacillus pumilus Spore stability in225.33×107CFU/mL, spore rate stable at around97.32%.
