OsCSLF6is a member of rice cellulose synthase-like family F and is essential for the synthesis of the (1,3;1,4)-b-D-glucans. In order to make further study of the function of OsCSLF6in cell wall, we obtain two oscslf6loss-of-function mutants and analysis the influence of oscslf6for plant type and cellulose synthesis in rice by molecular biology and cytology way.The main results are described as follows:1. Mutant phenotypes are both dwarf and less tiller in oscslf6-l and oscslf6-2lines. Co-segregation experiment shows the T-DNA insertion co-segregates with phenotypes. Complementation experiment indicates OsCSLF6is responsible for the plant height and tiller of mutant lines.2. Quantitative PCR analysis showed OsCSLF6gene is highly expressed in sheath, stem and flower at different development stages but lowly expressed in leaf. The mutation of oscslf6-1results in thinner secondary cell wall and vascular than wild type (WT) control. Further analysis revealed a20%reduction of cellulose content in oscslf6-1mutants than in their WT counterparts.3. The oscslf6-1mutants couldn’t be recovered to the WT phenotype by using hormone treatment with GA, ABA and BR. Co-expressing analysis showes that the expression of OsCSLF6is correlated to several other second wall genes such as cellulose synthase genes (CESAs). The real-time PCR indicates expression levels of several cellulose-related genes are changed in oscslf6mutants.
