Bryopsis hypnoides is a type of sea green alga that is present worldwide, which has a special repair mechanism. The extruded protoplasm can regenerate to new algal thalli in sea water, during this process, the cell is totally reproduced from the cytoplasm. Scanning electron microscope and fluorescence microscope was used to detect the regeneration process. Cytoskeleton was found involved in the regeneration and played a very important role. At the same time, a new method was developed to isolate the cell wall protein of Bryopsis hypnoides. Western blot was used to detect the contaminations in the cell wall protein samples. The result improved our method could extract cell wall proteins without cytoplasmic contamination. Two-dimensional electrophoresis was used to characterize the proteins and300protein spots were detected.47spots were chosen to take mass spectrometry detection. All47spots got MS signaling results and8of them got positive contrast in the NCBI database. The results of this study suggest complex signal regulation in the cell wall by proteins and enzymes. Thioredoxin-related lipid metabolism might play an important role in signaling and defense in the cell wall. B. hypnoides is a great material which can be used to study the cell wall. In the process of regeneration, cell wall formed from the very beginning that provide our a excellent opportunity to study the cell wall proteome.
