Based on HeLa as the research object, using conventional electron microscope thin slice technique and NAMA-Ur method to specific staining of nucleoli especially rDNA internal components, nucleolus structure observed under electron microscope in interphase cells and in the process of dynamic change of rDNA form. Through the research found that:1. After the synchronization of cultured HeLa nucleolus structure in interphase cells is different in each process. Several small nucleoli of G1phase in the cell nucleus, and a few larger FC is contained in each small nucleoli and DFC. Nucleolar volume become bigger, then formed a small number of large FC and DFC. S phase of the nucleolus in the region of the DFC form small lacuna, the lacuna were then developed into several small DFC and FC. In G2, accompanied by the FC and DFC atrophy and disappearance, nucleolus begins to collapse.2. Through the NAMA-Ur way to specific staining of rDNA, could eliminate the interference of other components in the nucleus. RDNA fiber by DFC in FC and FC stated coupon, nucleolus sec-FC derivative process occurs in primary DFC rDNA component of the lateral. Starting from G1phase, the DFC rDNA components as for increasing the cell cycle, until, late G2midterm nucleolus peripheral chromatin condensation began to set, in the bowels of the nucleolus chromatin gradually disappear, FC and DFC dyed lighter due to loss of rDNA components, nucleolus forms also disappear at the same time.
