Objective: Pre-implantation genetic diagnosis technology comes into being with fastdevelopment of both assisted reproductive technology and molecular biology, which has beenmore than20years of history. Pre-implantation genetic diagnosis is a good means to improvethe population quality, to control the birth defect and to treat infertility. Through theretrospective analysis of clinical cases of PGD/PGS, hope providing clinical resources forPGD/PGS technology widely developing and continuously perfect.Methods: During Nov2009to Mar2013the case information of pre-implantationdiagnosis and screening in Assisted Reproduction Center of Shaanxi Province is collected,and carries on the summary analysis. Pre-implantation diagnosis and screening methodsmainly include immune fluorescence in situ hybridization technique and array comparativegenome hybridization technology.Results: During Nov2009to Mar2013there are29PGD/PGS cases (31oocyte retrievalcycles) in the ART center getting477oocytes.403oocytes are in MⅡ; the total number offertilization is271; the number of cleavage embryo is265, the rate of cleavage embryo is97.79%; the number of2PN is254, the rate of2PN is63.03%; the number of high qualityembryo is104, the rate of high quality embryo is40.63%. The number of biopsied embryo is181; the number of successful biopsy is180; definite diagnosis embryo is171; the normal rateof the embryo is58.48%. Normal embryo detection rate using FISH is64.34%; using aCGHis28.57%. In the29cases, there are15cases of abnormal sex chromosome,11cases ofautosomal abnormality,3cases of recurrent miscarriage and1case of repeatedly failed IVF.In addition, there is one case in which the male is sex chromosome abnormality and thewoman is autosomal abnormality. The28transplanting cycles are carried out, and five boysand two girls have been born. Clinical pregnancy rate is42.86%; implantation rate is32.65%;abortion rate is25.00%. In fresh cycles the clinical pregnancy rate, implantation rate andabortion rate is26.67%,23.08%and26.67%respectively; In freezing cycle the clinicalpregnancy rate, implantation rate and abortion rate is respectively61.54%,61.54%and12.50%.Conclusion: Normal embryo detection rate using aCGH technology significantly lowerthan FISH; For a particular patient, PGS applications can serve as a kind of new way of embryo selection; After PGD, clinical pregnancy rate, implantation rate, abortion rate andfetal condition is satisfactory.
