Strain Mnl-7T was isolated from Tianjin manganese mining soil. Strain Mnl-7T was subjected to polyphasic taxonomic studies.Cells of Strain Mnl-7T are Gram-positive, aerobic, terminal spherical spore-forming, motile by lateral flagella and rod-shaped (0.3-0.7×1.7-2.5μm). Colonies are circular, convex, non-transparent, smooth and yellow. Temperature range for growth is15-45℃(optimum,37℃) and pH range is5.0-10.0(optimum, pH7.0). Cells grow in0-5.5%NaCl (w/v).This strain was negative reaction for nitrate reduction, arginine dihydrolase, citrate utilization, urease, Voges-Proskauer test, production of H2S and indole, and hydrolysis of casein and starch; positive for oxidase and catalase.The predominant menaquinone of strain Mn1-7T is MK-7. The major fatty acids (>10%) are iso-C15:0, iso-C16:0and iso-C14:0. The major polar lipids are diphosphatidylglycerol and phosphatidylglycerol. Cell wall peptidoglycan type was A4a (L-Lys-D-Asp) and the major cell wall sugar was xylose.The genomic DNA G+C content was38.4mol%. Analyses of16S rRNA gene sequences revealed that strain Mnl-7T was affiliated with the genus Lysinibacillus. The most closed relatives was L. massiliensis CCUG49529T (97.2%,16S rRNA gene sequence similarity). Strain Mnl-7T has the highest16S rRNA gene sequence similarities to the strain L. massiliensis CCUG49529T (97.2%), and94.1-96.7%sequence similarities to those of other recognized Lysinibacillus species. DNA-DNA hybridization values for strain Mnl-7T with L. massiliensis was24.9%.The results of the polyphasic taxonomy analysis revealed that the isolate represents a novel species of the genus Lysinibacillus, for which the name Lysinibacillus manganicus sp. nov. is proposed. The type strain is Mnl-7T (=DSM26584T=CCTCC AB2012916T).Identifications of new manganese ore strains will contribute to the study and exploitation of the manganese ore microorganism resources.
