4-phenyl butyrate (4-PBA) is a short chain fatty acids.4-PBA inhibites the activityof Histone deacetylase (HDAC), and reduces the response of cells to endoplasmicreticulum(ER) stress to improve glucose up-taking in diabetic mouses. The glucosetransporter is a essential factor in glucose up-taking. The purpose of this study was toinvestigate the effect of4-phenyl butyrate (4-PBA) on the glucose transporters geneexpression in C2C12myotubes, and to endicate the mechanism of glucose transporter4expression regulated by4-PBA, and approach the mechanism of diabetic pathologicalcharacteristics.To clarify the effect of4-PBA on the expression of Glut4gene expression inC2C12myotubes, Real-time PCR and western blot were used. In order to analyze thecorelation of ER stress with Glut4gene expression, the change of Glut4gene expressionin C2C12myotubes treated with tunicamycin (TM), an inducer of endoplasmicreticulum stress. After that C2C12myotubes were treated by butyrate, which is anHDAC inhibitor was measured. Then in order to investigate the correlation betweenHDAC5and Glut4transcription, C2C12myotubes were treated by butyrate, an HDACinhibitor, Glut4gene expression was detected.The result indicated that4-PBA increased gene expressions of Glut4in C2C12myotubes. While, there were almost no effects of increased ER stress response on geneexpression of Glut4. On the other hand, HDAC inhibitor butyrate promoted geneexpressions of Glut4in C2C12myotubes.4-PBA and butyrate also increased theacetylation of histone3, and diminished the activity of classâ…ˇhistone deacetylaseHDAC5.The results suggested that4-PBA enhanced gene expression of Glut4in C2C12myotubes.4-PBA could regulate the Glut4expression by suppressing the activity ofhistone deacetylase, but not by reducing endoplasmic reticulum stress.4-PBA may serveas a potential clinical reagent useful for treatment of diabetes mellitus.
