Objective To built a detection method of Chronic myelocytic leukemia.Methods The experiment at the based on the experiment of the lab’s selected5highly-combined aptamers of CML K562cells, constructed one detection methods: Marked five aptamers by using two kinds of nanoparticle,The ten marked aptamers respectively with the CMLK562combination alone, determination and record two metal content, The ten marked aptamers were divide20pairs,everyone pair combined with CMLK562cells,determination and record two metal content,To compare the two results,Each combination that the pair aptamers and K562cell with different immobilizations. The pair of aptamers, a as capture molecules, a determination as molecules combined with CMLK562cells, the molecules will determine into a state of metalions by the REDOX reaction,correspond current workstations by using electrochemical detection.Selected the best combination To detect the CMLK562Results1.Binding sites the results showed that five aptamers were mark by using two kinds of nano metal materials, the marked10aptamers with the CMLK562combination respectively, determination and record two metal content F e, Au content,the content were1.921ng/μL and1.352ng/μL. The ten marked aptamers were divided20pairs by combinating willfully two, The pairs combinated with CMLK562, determination and record two metal content, compare the two results. The results were no obvious difference (RSD=0.5211%,0.5865%), prove the binding sites of CMLK562and aptamers is not the same2.The best pair of aptamers was selected to detecte CMLK562, the fitted curve equation to test cells number and DPV of current value was obtainedConclusion Get a electrochemical biosensor which can be diagnosis of CMLK562cells,detection limit up to50cells...
