Three kinds of novel aptasensors were fabricated and investigated as followed:(1) The first aptasensor was fabricated by assembling thrombin aptamer and L-cysteineonto the Au electrode. The effect of experiment conditions of different pH value and theassembly time on the aptasensor were also studied. In the optimal experiment condition, theaptasensor was applied to detect thrombin and satisfactory results were obtained: the responserange is10ng/ml~10000ng/ml. What’s more, lgC (concentration) and the current value werein a linear relationship with a correlation coefficient of0.994in the whole response range.(2) A sensitive aptasensor was fabricated reling on the amplification of Pt nanoparticlesthrough the method of an improved sandwich. In the optimal experiment condition, theaptasensor was applied to detect thrombin and satisfactory results were obtained: the responserange is1ng/ml~1000ng/ml. Moreover, lgC and the current value were in a linear relationshipwith a correlation coefficient of0.99925in the range from1ng/mL to50ng/mL.(3) The the response range of the signal on aptasensor is1μg/ml~5μg/ml. In the wholeresponse range, lgC and the current value was in a linear relationship with a correlationcoefficient of0.991.
