Objective:To explore Intervention effects of aerobic exercise and clear-fat-herbal on high-fat-induced rats and its mechanismsMethods:60SD male rats were divided randomly into five groups:Control group (C group), High fat group (HF group), High fat diet+exercise group (HE group), High fat diet+clear-fat-herbal group (HZ group), High fat diet+exercise+clear-fat-herbal group (HZE group). The training plan for rats was60-min unloaded swimming which would be done6days per week for10weeks. The dose of Chinese traditional medicine was lml/100gBW (Density was0.8g/ml). By the end of experiment, the serum TG, TC, AST, ALT, LDH, FFA contents and liver TG, TC, SOD, MDA contents were detected, and also the expressing situation of PPARαmRNA by quantitative PCR testing method, while the morphological changes of liver tissues were observed under electric microscope.Results:1. Compared with C group, the serum TG, TC contents of HF group increased significantly (p<0.01), and the levels of TG, TC and FFA in the liver increased significantly, too (p<0.01). Through the double factor variance analysis, the aerobic exercise and clear-fat-herbal could both lower the serum TG,TC and FFA contents significantly in the high fat diet rats. Meanwhile, aerobic exercise and clear-fat-herbal displayed a obvious interaction on lowering TG, TC and FFA contents in serum and liver in high fat diet rats(p<0.01).2. Compared with C group, the serum ALT in HF group had a obvious trend of increasing (p<0.01), and serum AST and LDH activities also showed the increasing trend, and difference is obvious (p<0.01). Through the double factor variance analysis, aerobic exercise or clear-fat-herbal could lower the serum AST, ALT and LDH activities in HF rats significantly, and aerobic exercise and clear-fat-herbal have obvious interaction on lowering serum AST and LDH activities and reduce their activities further more.3. Compared with C group, the SOD activity in the livers of HF group decreased significantly (p<0.01), MDA content increased dramatically (p<0.01), so the ratio of SOD/MDA decreased remarkably (p<0.01). Through the double factor variance analysis, both aerobic exercise and clear-fat-herbal could increase the SOD activity in high fat diet rats’liver tissues(p<0.01) and lower the content of MDA significantly(p<0.05), so the ratio of SOD/MDA increased significantly(p<0.01). Aerobic exercise combined clear-fat-herbal can increase the SOD activity and decrease the MDA concentration in the liver of high fat diet rats, and showed a significant interaction effects. Compared with C group, The PPAR-amRNA concentration decreased significantly in the HF group (p<0.01)4. Compared with HF group, the PPAR-amRNA contents in the livers of HE and HZ group increased dramatically(p<0.01). Through the double factor variance analysis, aerobic exercise and clear-fat-herbal could both increase the PPAR-amRNA concentration significantly in the high fat diet rats(p<0.01), Aerobic exercise combined clear-fat-herbal could increase the PPAR-amRNA content in the high fat diet rats significantly, and showed insignificant interaction effects (p<0.5).5. In HF group, light microscope observation revealed swelling of hepatocytes and the liver displayed moderate and severe steatosis, different sizes of lipid droplets inside the cytopla sm. The hepato sinusoidal became narrowand there have been inflammatory. HE set of rat liver cells have normal, nuclear centers, a small number of vesicles, normal liver cable str uctures, sinusoidal clearance without noticeable change. The structure of hepato cell in H Z group were normal in principle, and the nucleolus of the hepato cell were in the center, f ew micro lipid vacuoles could be found. The hepatic cord tended to be normal and the hep ato sinusoidal became narrow slightly. The hepato cells in HZE group are basically arrang ed in a central vein center is radial, central nucleus is located in cells. Few microlipid vacu oles could be found in the cell occasionally. The structure of the hepatocable were normal and the hepato sinusoidal had no obviously change.Conculsions:1. Ten-week’s high fat diet not only successfully induced the rats to form non-alcoholic fatty liver disease, but also caused the formation of NASH.2. Concurrent with the formation of non-alcoholic fatty disease induced by high fat diet, aerobic exercise and supplement of clear-fat-herbal could effectively prevent high-fat-diet-induced fatty liver disease, further more, aerobic exercise and clear-fat-herbal displayed significant interaction in the formation of fatty liver disease.3. The oxygen free radicals increased roughly concurrent with high-fat-diet-induced fatty liver disease,it might be the mechanism of hepatocells damaged and the serum enzyme activities increased. Aerobic exercise and clear-fat-herbal could improve the metabolism of the free radicals in high fat diet rats to prevent the damage of liver.4. Concurrence with The formation of high-fat-diet-induced non-alcohol fatty liver, the rats’ lipid metabolism in blood and liver became disorder, which caused the transformation of PPAR-a mRNA contents in the liver, so provide new theory foundation of preventing and treating fatty liver disease.
