Objective:The objective of this study is to investigating the role and mechanism of IL-6in the training decrease blood pressure in spontaneously hypertensive rats and to exploring the roles and mechanism of prevention on hypertension and its complications in optimal exercise for providing theoretical basis for prevention and treatment of hypertension and its complications.Methods:The sixteen male Wistar rats were randomly divided into two groups:the Wistar control group and Wistar training group; The eighteen male spontaneously hypertensive rats were randomly divided into two groups:SHR control group and SHR training group;WT and ST groups carried on90minutes swimming6times every week during the experiment.Determined the rats blood pressure and the body weight each week during experimental period, determined indexes of protein contents、Ang Ⅱ、 NF-κB、IL-6and Ca2+of the vsmc in aorta。Result:(1) After the90minutes swimming every day for8weeks, we found the weight of Wistar training group more obviously descended than Wistar control group; the weight of SHR training group more obviously descended than SHR control group, It means that exercise can inhibit to the increase of weight in Wistar and SHR rats.The blood pressure in SHR control group more obviously raised than before experiment (P<0.01); the blood pressure of SHR training group although higher than before the experiment, that’s no significant difference; While the SHR control group, elevated blood pressure range of significantly reduced (P<0.01); the blood pressure of Wistar control group although higher than the Wistar training group, that’s no significant difference.It means that exercise can inhibit the increase of blood pressure in SHR rats but there’s no inhibition to blood pressure of Wistar.(2) After the90minutes swimming every day for8weeks, we found the Ang Ⅱ in vsmc of SHR control group are more than SHR training group (P<0.05); It means that exercise can inhibit the reduced of the Ang Ⅱ in vsmc by SHR rats; the NF-κB in VSMC of SHR control group are more than SHR training group (P<0.05); It means that exercise can inhibit the reduced of the NF-κB in vsmc by SHR rats; the IL-6in vsmc of SHR control group are more than SHR training group (P<0.05); It means that exercise can inhibit the reduced of the IL-6in vsmc by SHR rats; the Ca2+in vsmc of SHR control group are more than SHR training group (P<0.05); It means that exercise can inhibit the reduced of the Ca2+in vsmc by SHR rats.Conclusion:(1) After the90minutes swimming every day for8weeks can significantly inhibit the further increase of SHR blood pressure, and can mitigate the hypertension. Meanwhile in works on inhibiting the weight of Wistar and SHR rats from increasing;(2) After the90minutes swimming every day for8weeks can decrease the Ang Ⅱ amount of the aorta of SHR and Wistar, reduce the pressor effect of Ang Ⅱ,which plays quite an active role of preventing and reducing high blood pressure;(3) The experimental results show that IL-6in motion of vascular smooth muscle injury prevention and control hypertension plays an important role. The possible mechanism is that long time exercising regularly lower the content of Ang II in vascular smooth muscle cell of rats thus inhibiting vascular smooth muscle in the NF-κB activity expression. Reduce the inflammation of IL-6in vsmc to reduce the formation of lower phospholipid C activation degree, which through the excited-shrinkage reducing coupling mechanism Ca2+concentrations inside cells, improve the function of vascular smooth muscle cell, causes vasodilation reaction to play anti-hypertensie.
