Objective: To observe the effects of Polygonatumpolysaccharide on the low human T lymphocyte immune function caused byhigh-intensity exercise,and analyze the mechanism of the preceding changes,and todiscover new ways which may release the exercise-induced immune dysfunction.Approach:14boys who are no system of professional sports training experiencefrom sports university are randomly devided into placebo group(group A) andPolygonatum polysaccharide group(group B), each consisting of7persons. Thesubjects of group A take placebo(dextrin capsules) by the dose of1g/d, and thesubjects of group B take Polygonatum polysaccharide enteric-coated capsules by thesame dose. After respectively taking for4weeks, all subjects take one high-intensityincremental treadmill running, blood samples of10ml were taken at their cubital veinat the time before exercise, immediately after exercise and24hours after exerciserespectively. T-lymphocyte transformation idxe (T-LTI) was measured by the methodcalled MTT, the percentage of T lymphocyte subsets (CD4~+、CD8~+) was measured byflow cytometry, Th1,Th2-type cytokines(IFN-γ、IL-4) was measured by ABC-ELISA,plasma cortisol(Cor) was measured by radioimmunoassay(RIA). Results:1. Allindexes were no significant difference at the time before exercise between A and Bgroups.2. T-LTI of both groups at the time immediately after exercise were lower thanthe level before exercise, group A had very significant difference; at the time24hoursafter exercise, T-LTI of both groups were picked up, group A close to the pre-exerciselevel, slightly higher than before exercise level in group B; T-LTI of group B washigher than group A in each phases after exercise, there was significant differenceimmediately after exercise.3. CD8~+immediately after exercise were significantlyhigher than before exercise and CD4~+、CD4~+/CD8~+were significantly lower thanbefore exercise in group A, the three indexes have been restored24hours afterexercise, but did’t reach the pre-exercise level, CD4~+/CD8~+was still significantlylower than before exercise; the trend of the three index in group B were as the same asgroup A, but the change amplitude was significantly less than group A, there was nosignificant difference compared with before exercise, CD4~+、CD4~+/CD8~+restored toslightly higher than before exercise at the time24hours after exercise.4. IFN-γofboth groups at the time immediately after exercise were lower than the level beforeexercise, group A had significant difference, IFN-γof group B significant higher thangroup A at the time immediately after exercise, IFN-γof both groups were picked upto near pre-exercise level at the time24hours after exercise; IL-4of both groups at the time immediately after exercise were significant higher than the level beforeexercise, there were some decrease at24hours after exercise, but still higher than thelevel before exercise, group A had significant difference, IFN-γ/IL-4of both groupsat the time immediately after exercise were significant lower than the level beforeexercise, picked up at24hours after exercise, but still lower than the level beforeexercise, group A had significant difference.5. Cor of group A at the timeimmediately after exercise higher than the level before exercise, it still higher than thetime before exercise at24hours after exercise, but no significant difference, Cor ofgroup B at both immediately after exercise and24hours after exercise was lower thanthe level before exercise, but no significant difference. Conclusion: A high-intensityincremental treadmill running cause the suppression of human T lymphocyte immunefunction, pre-supplementary Polygonatum polysaccharide can reduce the decline of Tlymphocyte proliferation, CD4~+cell content and IFN-γlevel, and reduce the increaseof CD8~+cell content and IL-4level caused by exercise, thus prevent the decline ofCD4~+/CD8~+and IFN-γ/IL-4, indicating that Polygonatum polysaccharide caneffectively reduce the degree of immune balance tends to suppression and reduce thedegree of immune steady-state tends to humoral immune caused by exercise.Speculate the mechanism is that Polygonatum polysaccharide can inhibit the increaseof plasma cortisol after exercise, thus mitigate its negative regulatory effect onimmune function.
