Aim:(1) Impact of the different myocardial states on related acupoints skin blood flowTo comparatively observe the change of related and non-related acupoints skin blood flow in normal myocardial state, myocardial ischemia state, electroacupuncture(EA)-simulated state, it would find the changing law of the skin blood flow in different acupoints with the myocardial ischemia, and the specific effect of the skin blood flow changed on acupoints. Meantime, it would explore the law and feature of acupoints skin blood flow influenced by different frequency EA improving myocardial ischemia lesion.(2) Impact of the different myocardial states on related acupoints molecule(NOS/NO/CaM) related to sympathetic nerve(SN)To comparatively detect the change of related and non-related acupoints skin molecule(NOS/NO/CaM) related to SN in normal myocardial state, myocardial ischemia state, EA-simulated state, it would find the changing regularity of skin NOS/NO/CaM in different acupoints with the myocardial ischemia, and the specific effect of these molecule changed on acupoints. Meanwhile, it would explore the effect mechanism about the change of SN-related molecule(NOS/NO/CaM) in acupoints skin while regulating the process of myocardial ischemia by different frequency EA.Materials and methods:Fifty male Wistar rats with210-230g were randomized into normal control, sham-operation(used to detect skin blood flow), model, high frequency EA, and low frequency EA groups with10rats in each group. The electrocardiogram (ECG) of rats was detected with Ⅱ ECG, the time included before building the model, the instantly/10min/30min after building the model and the instantly/10min/30min after EA; skin blood flow was dynamically detected by Perfusion IMager (FIM). the time included before building the model, the instantly/10min/30min after building the model and the instantly/10min/30min after EA, the acupoints included"Neiguan"(PC6)."Ximen"’(PC4)."Tianquaif (PC2)."zusanli"(ST36), no-meridian-no-acupoint of lateral-above to ST362mm; skin tissue in acupoint area were extracted at last, CaM contents was detected by Enzyme-linked immunosorbent assay, the contents of NO metabolites nitrate NO-3and NO-2by Nitrate reductive enzymatic to indirectly reflect NO contents,NOS activity by Colorimetric method, the acupoints included PC6, PC4, PC2,"Waiguan"(SJ5). ST36. Results:(1)When rats occurred to myocardial ischemia, the D-value between J point and QRS initial point in the Ⅱ ECG was much bigger at30min after EA; the low frequency EA could effectively lower the D-value, and its instant effect was superior to that of high frequency EA, but the incubation period in the high frequency EA group was longer.(2)When rats occurred to myocardial ischemia, the skin blood Perfusion Unit(PU) of PC6and PC2in the left pericardium meridian in the model group were both increased significantly, while decreased significantly in the right stomach meridian acupoint ST36; the skin blood PU of PC6and PC2in the high frequency EA and low frequency EA groups both decreased tending to the normal level, and effect of the low frequency EA was superior to that of high frequency EA; in addition, the skin blood PU of PC4and ST36were decreased and increased respectively at10min after low frequency EA.(3)When rats occurred to myocardial ischemia, the NO contents and NOS activity of PC6, PC4and PC2in the bilateral pericardium meridian. SJ5in the exteriorly-interiorly related meridian, ST36in the stomach meridian all decreased: the CaM contents of PC6and PC2in the bilateral pericardium meridian and ST36in the stomach meridian all increased:after EA with high frequency, the NO contents of PC6, PC4and PC2all became increased; the NOS activity of PC6. SJ5and ST36all became enhanced; the CaM contents of PC6and PC2and ST36were decreased in the low frequency EA group, the CaM contents of PC2and ST36were decreased in the high frequency EA group.Conclusion:(1)After myocardial ischemia, the heart function was seriously affected with the longer about the myocardial ischemia time, the early effect of low frequency EA on improving the heart function was superior to that of the high frequency EA. but whose later effect was better:(2)the change of skin blood flow in the left related acupoints was manifested as the specific effect compared with that of the right stomach meridian acupoint ST36,high and low frequency EA of PC6affected acupoints skin blood How when protected from myocardial ischemia, manifested as the specific effect on frequency, and that of low frequency was better:(3)the change of NO contents and NOS activity of three acupoints in related pericardium meridians and SJ5in exteriorly-interiorly related meridians and ST36in high-power high-blood meridians were related to myocardial ischemia: then high frequency EA was superior to the low frequency EA manifested in the specific function of upregulating the levels;the change of CaM contents in PC6and PC2of related meridians and in ST36of high-power high-blood meridians were more related to myocardial ischemia, the levels all decreased after the high frequency EA and low frequency EA, so their difference was not significant.
