Purposes:To study the effects of Tetrandrine(Tet) on the proliferation and apoptosis of human triple-negative breast carcinoma HCC1937cell line in vitro.Methods:MTT was used to detect optical density(OD) of human triple-negative breast cancer cell HCC1937by intervention of Tet in seven different concentration groups in24h,48h,72h,96h. Drawing cell growth curve, calculating cell inhibitory rate and50%inhibitory concentration(IC50) in every group, which were used to show inhibitory effect of Tet on human breast cancer cell HCC1937in vitro growth.Fluorescence microscope was used to observe the morphology of cell apoptosis by intervention of Tet in three different concentration groups in48h.Cell apoptosis inspection method of AnnexinV-FITC, PI double labelling was used to detect the cell apoptosis effect of Tet on human breast cancer cell HCC1937in four different concentration groups after24h.The changes of Bax, Bcl-2and E-cadherin in protein levels were quantified by immunohistochemical method.Results:The growth inhibition of triple-negative breast cell was dose-dependent obvious after exposure to the Tet which was divided into different concentrations groups. With the Tet concentrations rising and the period lengthening, the more increasingly the Tet effected. All the treatment groups were observed much more early apoptosis cells than the control group under the fluorescence microscope. The rate of early and late apoptosis in24h was increased in four different Tet concentration groups. Significant positive correlation was observed between apoptosis rate and Tet concentration. With the Tet concentrations rising, the expressions of E-cadherin protein was significantly increase.There are no significantly difference in expressions of Bax and Bcl-2protein.Conclusion:The Tet was proved to be effective in the inhibition of the proliferating and the induction of the apoptosis of human triple-negative breast carcinoma HCC1937cell line and with the Tet concentrations rising and the period lengthening, the more obviously the Tet treated. Tet can induce early and late apoptosis of HCC1937cell, and the effects has positive correlation with the concentration of Tet. There would be some other mechanism of pro-apoptosis on HCC1937cell to Tet, because Tet has no obvious effect on the expressions of Bax and Bcl-2.But the result, that increased expression of E-cadherin related with Tet concentration provide foundation to the studies on how Tet regulates the transfer and recurrence of triple-negative breast carcinoma.
