Part Ⅰ Clinical researchObject: This study focused on the investigation of the symptoms of asthma patients, and analysis of the related data Cor statistical, to explore the syndrome characteristics of bronchial asthma.Method: Select83cases from September2012to February2013of the diagnosis oC bronchial asthma, by the analysis of symptoms of bronchial asthmaResults: Dysfunction of five internal organs can lead to asthma, while all the83cases show the dysfunction of lung,39cases are related with liver,34with spleen,30with kidney,27with heart. Stable asthmaIic shows notable syndromes performance compared wi th the acute attack stage. The longer the duration lasts, the more organs deficiency syndrome type? performance tends to be significant, the number of cases increase significantly.40%of patients in patients with bronchial asthma is also diagnosed in patients with allergic rhinitis; The patients taking systemic glucocortieoid tend to temperament changing, P<0.05.Conclusion: Further confirmation the important role of. dysfunction of liver in the onset and aggravating process of asthma, providing theoretical support for the treatment of bronchial asthma from the liver.Part Ⅱ Experimental studyObject: To explore the clinic effect of Xiaochuanning Granules on bronchial asthma, medicated serum of Xiaochuanning Granules was prepared in rat, the anti-degranulation effect on rat basophile leukemia cell line(RBL-2H3) was observed in vitro.Methods:30adult Sprague-Dawley rats weighing between180g-200g were randomly divided into5groups of five rats each. Animals in group A received physiological saline orally and served as control while hexadecadrol treated rats (group B) received dose of8.33μg/ml, as positive control by gavage. Xiaochuanning Granules treated rats (group C, D,E) were received different doses of40,20,10mg/mL respectively for5continuous days. Medicated serum was separated; anti-degranulation effect was observed with5%and10%medicated serum respectively. The real time change of intracel1ular calcium was recorded by Calcium Imaging Method and the release of β-hexosaminidase was detected by colorimetric method.Results: The medicated serum of Xiaochuanning Granules can inhibit both the early and late phase of degranulation after sensitized by Compound48/80by the reduction intracellular calcium and,β-hexosaminidase release.Conclusion:Medicated serum contains the metabolic components of Xiaochuanning Granules. The results highlight the curative effect of Xiaochuanning Granules on allergic asthma and bronchial asthma in clinic might be a possible result from the anti-degranulation effect.
