Depression is a kind of mental disorder that takes notable long-term low spirit as the main manifestation. It is frequently complicated with anxiety, insomnia, and the feeling of guilt. The disease features in high incidence, recurrence, and suicide rat. And it is of low awareness and cure rate. The disease, frequent attacking the old and adolescents, and causing notable loss in economy, makes it one that worth studying.By now, modern medicine has assured that the disease concerns pathological changes of the hippocampus and the frontal lobe. It is likely to be caused by increase of inflammatory factors. Treating depression by western medicines has comparat ively great s ide effects and low compl iance. On the contrary, acupuncture therapy shares the same curative effect as western medicine do, and can greatly improve patient’s memory and anxiety. It is therefore considered as a possible treatment of depression. With its curative effect approved in clinical observations and animal experiments, its mechanism requires further study.Object ives:The research is to observe interference of depression model animals by different kinds of moxibustion therapies, including moxa smoke inhaling, point stimulation, and heating, to find out the mechanism of moxibustion therapy.Methods:40Male SD rats were divided into5groups randomly:blank group, model group, moxibustion group, moxa smoke inhaling group, and smokeless moxibustion group. All groups except blank group underwent chronic unpredictable moderate stress to set up models. After that, animals in moxibustion group was intervened with over skin moxibustion. Animals in moxa smoke inhaling group inhale moxa smoke of a potency of30mg/m3. And animals in smokeless moxibustion group"was intervened with over skin moxibustion without taking into any moxa smoke. At the end of the treatment course, check changes of the results of open field test, sucrose consumption, the contents of IL-1β and TNF-α in rats’blood serum, and that of NF-κB in rats’ hippocampus.Resulis:(1) Compared wi th the blank group, animals in the model group have reduced average moving speed and longest single movement (P<0.05). The differences can be reduced in animals of all the three treatment groups after the treatment course.(2) Hematoxylin and eosin stain showed that the cells are not in compact arrangement of blank group. There’s obvious damage of neuron cells in hippocampus area of model group animals. And the nucleus disappeared in some of the cells. In comparison, the number of neuron cells of hippocampus area was significantly increased and the form of cells is greatly improved in all the three treatment groups.(3) The content of IL-1β and TNF-α in blood serum of the model group animals is increased compared with that of the blank group (P<0.05). While the indices was reduced on animals in the three treatment groups (P>0.05).(4) Real-time PCR results shows that there’s a significant decrease in NF-κB in hippocampus area of the model group, moxibust ion group and smokeless moxibustion group animals compared with that of the blank group (P<0.01). The excess is greatly decreased in the moxa somke inhaling group (P<0.05).Conclusion:1. Moxibustion therapy has certain curative effect on depression, especially on behavior of depression animal. Moxa smoke inhaling, heating, and point stimulation have specific influence on animal model of depression.2. Different kinds of moxibustion therapies take effects on different indices. Moxa smoke inhaling, heating, and point stimulation may interfere depression in different ways.
