| Baishao originates from the dry root of Paeonia tacti lora Pall.There are three authentic regions,including Hao Baishao from An Hui,Hang Baishao from Hang Zhou and Chuan Baishao fromSichuan.Among them,Hao Baishao from An Hui has a high production and good quality,therefore,HaoBaishao is the main product of Baishao medicinal materials recently.We used Hao Baishao for this research project.Baishao,as well as its prepared and precessed goods,are widely applied in the clinic,and they play an important role in various preparations.The Baishao preparations recorded in Chinese Pharmacopoeia(2010) only includes Chao Baishao(fried) and Jiu Baishao(prepared with wine),however,with the development of modern preparation technologies,adopting the mew modern fermentation processing technology in Baishao,we obtain to get a new type of Baishao processed variety.Doing primary and systematic comparative research on the chemical ingredients of fermented Baishao.replacing traditional Baishao with this processed Baishao in the common formulas in clinic,based on the physical and chemical properties of each herb in the formula and pharmacology and medicinal effect study.combining the requirements of clinic application of herbs,making the dosage form choices from the formulas,we determined Gu Shang Ling Cha Ji,and did the research on preparation process,quality standard and primary stability.Making the comprehensively comparative research of the chemical constitution of fermented Baishao,Baishao medicinal materials and other preparations(heating with bran or prepared with win):â‘ Mmake the integral chemical composition analysis by using thin layer chromatography;â‘¡Test the contents of paeoniflorin in Basishao medicinal materials and other different preparations by means of High Performance Liquid Chromatography (HPLC)and then do the comparative analysis;â‘¢Do the comparative research on finger-print of Basishao medicinal materials and other different preparations;â‘£Make the analytical research on the chemical composition of petroleum ether from Basishao medicinal materials and other different preparation by gas chromatography-mass spectrometry,moreover,it is the first time to use this technology to study the petroleum ether of Basishao. Gu Shang Ling Cha Ji is processed from Yin Yang Huo,Bai Shao.Xu Duan,Da Huang and other medicinal materials.Based the physical and chemical properties of its effective ingredients,applying modern preparation processing,combining the clinic application necessities,choosing after multi-index comprehensive investigation,we finally determined the processing parameter of Gu Shang Ling Cha Ji.And after pilot production verification,it is proofed to be reasonable,stable and feasible.In the quality standard research part,we mainly did the research on raw material quality standard and preparation quality standard.In the raw material quality standard,we did various examinations on the pieces of the medicinal materials included according to Chinese Pharmacopoeia(2010).In the preparation quality standard,we did the linimentum general exam and qualitative discrimination of three batches of samples producted by pilot according to the Pharmacopoeia,and tested the content of icariin by HPLC,thus to control the content of the samples.We also established thin layer identification method of each formula medicinal materials of the related reference substances and referred medicinal materials in the complex preparations; tested the content of icariin and did the index composition control.Comparing the exam results from the three batches of samples,it shows that the transport rate are all more than60%,which states that there is only few loss in the processing procedure.Through storage observation of the three batches of samples under normal temperature,the results show that the stability of Gu Shang Ling Cha Ji is quite good. Applying fermented Baishao into preparations,we can save the amount of herbs used,so as to improve the availability of Baishao.Moreover,for the sake of the generation of new active parts,the clinic curative effects can also be improved.Our future task will be focusing fermented Baishao to further explore the chemical composition, pharmacology and medicinal effect completely and systematically,and do the evaluation on effectiveness and safety of fermented Baishao and its preparations. |
