Objective: Through the study of the Gexia-zhuyu Decoction combined with weste-rn medicine of commonly used chemotherapy drug cyclophosphamide (CTX) on EAC mice with hepatocellular carcinoma tumor interstitial vascular number, angiogenesis, vascular endothelial growth factor (VEGF). microvessel density (MVD) and other aspects,to explore its anti-hepatoma effect mechanism.Material and method:Take50kunming mice, according to the body weight completely randomized into blank group, model group, Chinese medicine group, Western medicine group and Combined drug group. Under the sterile conditions, in addition to the blank control group, the remaining mice in each group were inoculated subcutaneously EAC mouse hepatoma cell line suspension(2.0×106/ml,0.2ml/single) in the left axillary. After48hours, the mice were given normal sal ine (the blank control group), saline (the model group), Gexia-Zhuyu Decoction, CTX, Gexia-Zhuyu Decoction combined with CTX, continuous administration of ten days to observe the general status of the mice in each group, detected the autonomic activity and body weight, observed the number of tumor interstitial vascular by light microscopy and the tumor interstitial angiogenesis by electron microscope, tested the expression of vascular endothelial growth factor (VEGF) and intratumoral microvessel density (MVD) by immunohistochemical.Results:1. The general state of mice in each group:Mice in model group, spirit is di spirited, eating less, color dull and slow movement;western medicine group, the mental state of mice was bad, eating has increased than those in the model group, hair color is darker and the movement was smooth;in the Chinese medicine group, the spirit of mice was poor, the food intake compared with model group was increased,fur was smooth, slightly slow motion;combined drug group,mice were in good spirits, food intake increased significantly compared with the model group, fur was smooth and movement was more flexible, close to the general state of normal mice.2. The autonomic activity of mice in each group:Compared with the model group, the number of standing and independent activity of mice in Chinese medicine group, western medicine group and combined drug group was increased in five minutes, and the combined drug group was more close to the blank control group, there was no significant difference in the two groups (P>0.05).3. The body weight of mice in each group:Compared with the blank group, the weight of mice in others decreased significantly, and the model group lost the most; compared with the model group, the body weight of mice in Chinese medicine group, western medicine group and combined drug group have different degrees of growth, and taking combined drug group on mice weight growth was most close to the blank group.4. The number of mice tumor interstitial vascular in each group:Under the light microscope, the number of visible capillary was6-12at high magnification view (200times) averagely in the tumor stroma of the model group, which was intensive than those in the Chinese medicine group, western medicine group and combine drug group;and in the tumor stroma of the combined drug group, the average high magnification view (200times) only had3-5capillaries, as the minimal number of newly born capillaries group.5. The tumor interstitial angiogenesis of mice in each group:Under the electron microscope, within the tumor tissue in model group, the new capillary tube wall was thin, the gap between endothelial cells was big and basement membrane was thin and loose. And in Chinese medicine group and combined drug group, new capillary tude wall was thin and discontinuous, endothelial cells in a continuous contact between the pseudopodia could be seen part of the remaining fragments of blood vessels, but the combined drug group, but taking combined drug group of vascular structural damage was most severe.6.The changes of mice in VEGF and MVD:Immunohistochemistry showed that the positive expression of VEGF and MVD values in the Chinese medicine group, western medication group and combined drug group were lower than those in the model group, and the combined drug group reduced most.Conclusion:1. Gexia-Zhuyu Decoction combined with cyclophosphamide could improve the gener-al state of the EAC hepatocellular carcinoma in mice and enhance the quality of their life.2. Gexia-Zhuyu Decoction combined with cyclophosphamide can inhibit the EAC liver stromal tumor angiogenesis.3. Gexia-Zhuyu Decoction combined with cyclophosphamide can reduce the EAC liver cancer tumors in mice the role of VEGF expression.4. Gexia-Zhuyu Decoction combined with cyclophosphamide can reduce the EAC liver cancer tumors in mice the role of MVD value.5. Gexia-Zhuyu Decoction combined with cyclophosphamide can improve the quality of life of the EAC liver cancer in mice, inhibiting tumor stromal blood vessel growth may be achieved by reducing the role of VEGF protein expression and reduced tumor interstitial capillary density achieved.
