Luteolin, a representative natural flavone, that occur ubiquitously in plant kingdom, hasbeen shown to have biological activities such as antioxidant, antibacterial, antineoplastic et al. Inthis paper two complexes were synthesized by reacting luteolin with tow metal elements, Zn(II)and Cr(III), respectively to enhance the pharmaceutical activities. And the synthesizedcomplexes were characterized by a series of characterization means. Then their antioxidantactivities and inhibitory effects for α-glucosidase were investigated.In the synthesis reaction, we determined the synthetic methods of the two complexes, thencalculated the yields of the complexes. The complexes were characterized with the methods ofIR-spectra, UV-spectra, TG-DAT and element analysis. Then free radical (·OH, O2·-, DPPH·)scavenging activities of luteolin and the complexes were studied respectively by using threemethods and DPPH free radical scavenging activities were investigated respectively by usingHPLC and UV-spectrophotometer. Inhibitory effects of luteolin and the complexes on α-glucosidase were studied by using UV-spectrophotometer. And the resules were following:(1) Zn-complex was prepared by reacting luteolin with Zn(II) in ph8.7~9.8ethanolsolution in the temperature of80℃for four hours, and the yield was68.76%. Cr-complex wasprepared by reacting luteolin with Cr(III) in ph9.2~10.0ethanol solution in the temperature of80℃for four hours, and its yield was72.53%.(2) Compositions of the complexes wereC15H9O6Zn(COOCH3)(H2O)·H2O and C15H9O6Cr(COOCH3)2(H2O)2·2H2O respectively.(3)The IC50of·OH free radical scavenging activities of luteolin, Zn-complex and Cr-complexrespectively were0.288mmol·L-1,0.262mmol·L-1and0.173m mol·L-1.(4) The IC50of O2·-freeradical scavenging activities of luteolin, Zn-complex and Cr-complex respectively were0.355mmol·L-1,0.347mmol·L-1and0.299mmol·L-1.(5) The IC50of DPPH free radicalscavenging activities of luteolin, Zn-complex and Cr-complex respectively were1.46×10-5mol·L-1、1.41×10-5mol·L-1and2.33×10-5mol·L-1by using UV. And the IC50of DPPH freeradical scavenging activities of the three compounds respectively were4.92×10-5mol·L-1、0.682×10-5mol·L-1and1.52×10-5mol·L-1by using HPLC.(6) The IC50of inhibitory effects ofluteolin, Zn-complex and Cr-complex on α-glucosidase respectively were1.50mmol·L-1、2.52mmol·L-1and0.232mmol·L-1. The results showed that free radical scavenging activities of the complexes on·OH andO2·-were stronger than that of luteolin. And DPPH free radical scavenging effect of Zn-complex was stronger than that of luteolin and Cr-complex by using UV. But, DPPH freeradical scavenging effects of the two complexes were stronger than that of luteolin by usingHPLC. The results of inhibitory effects for α-glucosidase showed that the complexes werestronger than that of luteolin.
