Objective:This study was conducted to explore the feasibility and potential value of deuteporfin in tracing the metastatic lymph node of pancreatic cancer.Methods:(1) SD rats, according to the method of deuteporfin administration, were divided into two groups:caudal vein administration group and foot pad administration group. At1,3,6,12,24and48h after administration respectively, some of SD rats were sacrificed to harvest the heart, liver, spleen, lung, kidney, pancreas, popliteal lymph nodes, partial skin and blood for examination. We measured the concentration of deuteporfin in the various removed organs of the rats by fluorometric method and compared the effects of two administration methods on the distribution of deuteporfin in the body of animal.(2) By using BxPC-3cell lines and by applying continuous in vivo screening method, we prepared highly pancreatic metastatic lymph node models in nude mice. Nude mice were divided into normal nude mice group and model nude mice group. At1,2,4,6,12,24and48h after the caudal vein administration of deuteporfin, nude mice were sacrificed to harvest the bilateral popliteal lymph nodes. Concentration of deuteporfin in extracted lymph nodes was measured by fluorometric method and difference of deuteporfin uptaken between normal popliteal lymph nodes and metastatic popliteal lymph nodes were compared.(3) Wood’s lamp (wavelength400nm) was used to observe the photosensitive tracing phenomenon of lymphatic metastasis of pancreatic cancer that deuteporfin brings.Results:(1) Deuteporfin can get into the lymphatic system through peripheral blood administration.(2)24h after caudal vein administration, the concentration of deuteporfin was44.6ug/L in the metastatic lymph nodes, significantly higher than that (17.3ug/L) in the normal lymph nodes.(3) The differences of drug concentration can be expressed as fluorescence intensity, which can be identified by naked eye. Deuteporfin can produce fluorescent tracing phenomenon with light excitation at wavelength of400nm.Conclusion:(1)Deuteporfin could get into animal’s lymphatic system by vein injection.(2) Deuteporfin could be uptaken by metastatic lymph nodes selectively and has certain targeting ability to tumor tissue.(3) At excitation of400nm wavelength light, deuteporfin can produce visible fluorescence, which has a significant role in tracing. Deuteporfin-mediated photosensitive tracing effect is expected to be a new method of tracing metastatic lymph node.
