BackgroundHepatocellular carcinoma (hepatocellular carcinoma, HCC), is one of the common malignant tumors in China, the mortality rate of malignant tumors is second in China. The preferred method for the treatment of primary liver cancer is surgical resection, however, previous studies shown that the rate of3-year recurrence hepatocellular carcinoma and metastasis is as high as57%-81%, a serious threat to the health of people’s lives. Liver cancer was highly malignant and early prone to recurrence and metastasis, is closely related to their biological behavior. Many scholars have done a lot of research on the biological behavior of HCC found to play an important role in DNA damage repair in the liver during the development of multiple tumor suppressor genes, oncogenes, and protein regulatory factors in DNA damage play an important role in the repair process, tumor suppressor genes are a class of inhibition of cell over-growth, proliferation, and thus to contain the genes of tumor formation. The tumor suppressor gene tumor suppressor function by preventing cell cycle control can become unrestricted proliferation or by inducing tumor cell apoptosis. Loss or mutation of tumor suppressor gene is one of the reasons that lead to tumorigenesis. Zhao Xintai, etc. transfer HCCS1gene into liver cancer cells significantly inhibited tumor cell cloning HCCS1formation compared with control group,the in vitro studies confirmed the important role of HCCS1gene in hepatocellular carcinoma.Our studies focus on chromosome17p13.3region that are high-frequency loss of heterozygosity HCCS1(HCC suppressor gene1) gene in hepatocellular carcinoma and analyze its relationship with clinical factors.ObjectiveThis project aims to study HCCS1expression in clinical samples and to analyze its relationship with clinicopathological parameters and prognosis, as evidence of the tumor suppressor gene.Meterial and MethodsBy using Immunohistochemical staining (EnVision method),we compared the expression between hepatocellular carcinoma and paraneoplastic in tissue level (level two scoring method) using pearson correlation test liver cancer tissue HCCS1the correlation between the expression and clinicopathological parameters, compare HCCS1expression (positive and negative) relationship with prognosis and follow-up data using the Log-rank method for log-rank test and Kaplan-Meier method for survival curves, Cox proportionalhazard model multivariate analysis of survival.Results1. We detected of the expression of the24cases of paraffin-embedded tissue HCCS1by immunohistochemistry, HCCS1protein expressed mainly in the cytoplasm, compared to the lack of a majority of HCC tissues expression, and adjacent noncancerous liver tissue is usually positive (positive rate91.6%); t-test results showed that cancer and paraneoplastic expression difference was highly statistically significant (p<0.05); homogeneity of variance test next carcinoma and an independent sample t-test results indicate the data between the two groups from two normal distribution of samples, homogeneity of variance, comparable, support the results of the paired samples t-test.2. By compared HCCS1expression with clinicopathological parameters (including age, tumor size, TNM stage, pathological grade) line pearson correlation test, which HCCS1expression was highly negatively correlated (P<0.01) with hepatitis B history.3. Univariate analysis showed that HCCS1expression and prognosis (P<0.05), multivariate survival analysis showed that the prognosis of patients with hepatocellular carcinoma with tumor size, TNM stage, history of hepatitis (P <0.05), HSSC1expression significantly not correlated (P>0.05).ConclusionThe study found that the expression of tumor suppressor genes HCCS1in cancer tissues and adjacent tissues of liver cancer there is a big difference, detection of HCCSl may early diagnosis and prognosis of hepatocellular carcinoma. Tip we can take advantage of the tumor suppressor gene HCCS1, the use of different gene transduction vectors for gene therapy of liver cancer. HCCSl plays an important role in the induction of apoptosis and cancer treatment, may become a new molecular target for antitumor therapy.
