Matteuccia struthiopteris (L.)Todaro is a herbaeeous plant in Onocleaceae Matteuccia Todaro. M. struthiopteris are distributed over the margins of forests and the streams. M. struthiopter foliage leaves are rich in a variety of protein, vitamins, polysaccharides and other nutrients. Its rhizomes are rich in a variety of physterons, which has high development and utilization value, such as edible, medicinal, ornamental. In this paper, total physterone and antioxidant activity in the different regions and different periods of M. struthiopteris was determinated, the chemical composition and pharmacological activity were evaluated comprehensively, in order to provide the theory and experiment basis for the development and utilization of M. struthiopteris. The experimental conclusions are as follows:(1) We optimized the extraction and detection wavelength of total physterone in M.struthiopteris, and determined the optimal detection wavelength as570nm. The optimal extraction method is accurately weighed Matteuccia struthiopteris powder and join the60%ethanol by the ratio of raw materials to solution of40:1, ultrasonic extracted50min under the conditions power of270W. The filtrate vacuum evaporated with methanol to volumetric.(2) We determined and compared the total physterone content of the three kinds of Matteuccia plants, and the results showed that M. struthiopteris is the highest and has the significant difference, compared with M. orientalis(Hiook.) Trev. and M. intermedia C. Chr.. The highest content is in September with very significant difference in M. struthiopteris foliage leaves; The highest content is in Xunyang dam region with very significant difference in M. struthiopteris compared with other regions, and the content in the rhizomes and the foliage leaves were significantly higher than in the sporophyll.(3) We determined and compared antioxidant activity in vitro of M. struthiopteris ethanol extraction in different areas and different acquisition time, and the results showed that M. struthiopteris ethanol extraction has high antioxidant activity. Different medicinal parts of the extraction on DPPH radical scavenging capacity is:the rhizomes> the foliage leaves> BHT> the sporophyll; compare different parts of the scavenging capacity of the strongest is the Xunyang dam, as the harvested time extension, the scavenging activities is increasing,the strongest scavenging activity of the sample in September.The antioxidant capacity of beta-carotene/linoleic acid system assay showed that the samples have antioxidant capacity in180mins, no significant difference between each sample. Fe3+reducing power system assay showed that the Matteuccia struthiopteris extract antioxidant capacity gradually increased with the increasing of the reaction concentration, the sample antioxidant capacity higher than the BHT, when the reaction concentration is higher than1.0mg/mL, the antioxidant capacity of M. struthiopteris is strongest in Xunyang dam. Comparing with different acquisition time consider the strongest antioxidant activity of the sample in September. Comprehensive comparison the antioxidant activity in these three anti-oxidation system, the antioxidant capacity of M. struthiopteris is strongest in Xunyang dam. September have the best antioxidant activity.(4) We determined the levels of T-AOC, T-SOD and MDA in liver tissue and blood serum of mice, and studied the antioxidation effect in vivo of the rhizome from M. struthiopteris. The results show that the Mouse liver tissue the T-SOD activity, MDA and T-AOC and the normal control group have a significant difference, when the quantity of administration is50mg/kg/d, indicating that the Matteuccia struthiopteris extract can significantly improve mouse liver tissue the antioxidant capacity. The indicators the serum test results showed that give drug50mg/kg/d, T-SOD activity, MDA content and the normal control group compared have significant difference, give drug200mg/kg/d the total antioxidant capacity (T-AOC) with the normal control group have significant differences. This study showed that Matteuccia struthiopteris rhizomes can effectively improve the mouse ability to scavenging oxygen free radicals, thereby reducing by oxygen free radicals attacks to the mice cell cause damage, effectively raising the antioxidant capacity of the mouse.
