BackgroundAdipose tissue-derived stem cells (ADSC) and mature adipose can be isolated from adipose tissue. Mature adipocytes have been shown to possess the abi1i ty to lose the fat and revert to a stem-eell-ike phenotype, so-called dedifferentiated (DFAT) cells. The DFAT and ADSC have been demonstrated to he able to generate cardiomyocyte-like cells.Oxytocin, a female hormone, has been discovered to have the function of promoting cardiogenesis. It was reported that under the action of oxytocin P19and P19CL6cells could differentiate into cardiomyocytes. Although the precise mechanism of the effect of oxytocin is not clear, oxytocin may play an important role in the differentiation into cardiomyocytes from primitive cells including adult somatic stem cells.PurposeThe aim of this study is to explore whether Oxytocin (OT) could induce cardiomyogenic differentiation of ADSC and DFAT in vitro.MethodWe isolated ADSC and mature adipocytes from human adipose tissue. Mature adipocytes dedifferentiated into DFAT via ceiling culture. ADSC and DFAT were cultured with OT (10-8M,10-7M,10-6M) for1week,2weeks,3weeks in vitro. ADSC and DFAT cultured without OT served as control. Cells were evaluated morphologically and immunocytochemically, as well as by RT-PCR and Western Blot. Human cardiomyocytes were used as positive control.ResultThe majority of ADSC and DFAT treated with high concentration of OT(10-6M) died in the first two clays, and the subsequent experiment s were canceled. After treated with oxytocin (10-8M,10-7M) for3weeks, the ADSC and DFAT eells expressed cardiac marker including Nkx2.5, GATA4, cardiac troponin T on both gene and protein level. However, no spontaneous beating phenomenon was observed.ConclusionWe concluded that oxytocin (OT) could induce cardiomyogenic differentiation of ADSC and DFAT.
