Objective:To compare the performance of three kinds of dye solution on different concentration in micro leakage detection experiments of obturator, by measuring the depth of their penetration in the edge of obturator.Methods:Ninety recently extracted human second premolars teeth were chosen and randomly assigned to9groups (A1、A2、A3、B1、B2、B3、C1、C2、C3, n=10). Those teeth were cut on the buccal surfaces with the gingival margins placed1.0mm above the cemento-enamel junction (CEJ), then Class V cavities (4mm x3mm and2mm deep) were standardized mixed. Afer conventional resinous fillings, they were thermocycled for400cycles at5℃and55℃, sealed with nail varnish except1mm beyond the margins of restoration twice, and then separately immersed in different dye solution for96hours (A1, fuchsin solution,0.5%; A2, fuchsin solution,1%; A3, fuchsin solution,2%; B1, methylene blue solution,0.5%; B2, methylene blue solution1%, B3, methylene blue solution2%; C1, gentian violet solution,0.5%; C2, gentian violet solution,1%; C3, gentian violet solution,2%). Samples were then sectioned mesiodistally and viewed at40x magnification under a root canal microscope for leakage at the gingival margin and taken pictures. The microleakage was measured with the image analysis software Image-Pro Plus6.0and the data were recorded in data of dye penetration to the whole path from the cavosurface margin of the proximal boxes to the end of the post. Analysis of variance and the T-test were used to evaluate the data (p=0.05).Results:The depth of the penetration of dye solution of the nine groups of teeth separately were Al(0.72±0.55)mm、A2(1.25±0.35)mm、A3(0.93±0.24)mm、 B1(0.74±0.33)mm. B2(1.51±0.52)mm、B3(0.96±0.28)mm.Cl(0.59±0.22)mm、C2(1.38±0.32)mm、C3(1.52±0.45)mm. There were significant differences among the groups of A1,A2and A3in the depth of the penetration of fuchsin solution (F=4.570, P<0.05). And there were significant differences between A1and A2(t=2.881, P<0.05),also between A2and A3(t=2.234, P <0.05). But not between A1and A3(t=0.647, P>0.05). There were significant differences among the results of the groups of B1, B2and B3(F=10.398, P<0.05). And there were significant differences between B1and B2(t=4.443, P<0.05),also between B2and B3(t=3.155, P<0.05). But not between B1and B3(t=1.287, P>0.05). There were significant differences among the results of the groups of C1, C2and C3(F=21.431, P<0.05). And there were significant differences between C1and C2(t=5.138, P<0.05), also between C1and C3(t=6.082, P<0.05). But not between C2and C3(t=0.944, P>0.05).Conclusion:①In microleakage detection experiments of obturator, in which we used fuchsin solution, the penetration speed of fuchsin solution of1%had the biggest speed. That of2%and0.5%was a little slower. The penetration stability of fuchsin solution of2%was the best, that of1%was a little worse, that of0.5%was the worst.②In microleakage detection experiments of obturator, in which we used methylene blue solution, the penetration speed of methylene blue solution of1%had the biggest speed. That of2%and0.5%was a little slower. The penetration stability of methylene blue solution of2%was the best, that of0.5%was a little worse, that of1%was the worst.③In microleakage detection experiments of obturator, in which we used gentian violet solution, the penetration speed of gentian violet solution of1%and2%had bigger speed. That of0.5%was the slowest. The penetration stability of gentian violet solution of0.5%was the best, that of1%was a little worse, that of2%was the worst.
