Hypoxia Induced Glioma Stem Cells To Endothelial Cell Differentiation Potential Of Experimental Research

Objective To investigate the potency of glioma stem cells differentiate into vascular endothelial cells and participate in the formation of tumor vascular. Methods GSCs were extracted from glioma U251 cell lines using serum free medium and were induced to differentiate in the endothelial cells medium. CD31 CD34 were indentified after differentiated with the method of immunofluorescence technology. We implanted the post-induced cells into Matirgel medium, and observe its morphology character. We primary cultured the normal umbilical vein endothelial cells and made it as a contrast with the pre-induced and post induced GSCs, NO in the medium of the three parts were measured with the chemical enzymatic method and compared with each other. Results GSCs can be differentiated into VECs in vitro and express endothelial cells special marker CD31(+) and CD34(+). It can form tuber-like structure in Matrigel medium and secrete NO witch displayed part function of mature endothelial cells. Conclusion GSCs can be differentiated into VECs and participate in tumor vascular formation.
