Cotton Nicotinamide Gbnocotin Synthetase Gene And Preliminary Analysis Of The Promoter Function

The lack of Iron and B vitamins affects human health. Nicotianamine synthase (NAS) plays a key role in responses Fe-deficiency in graminaceous monocots. Nicotinamide (NA),synthesized by NAS, acts as an important carrier in the internal transport of Fe2+in many higher plants.iron and other divalent metal ion uptake and transport of an important carrier. In addition, nicotinamide (NA), also known as vitamin B3, plays an important role in the treatment of human skin diseases.In this paper,based on the previous research of our lab, a new NAS gene GBNocotin and its transcription factor gene were isolated from Gossypium barbadense by PCR, The main results of study are as follows:1. The results of bioinformatics analysis to GBNocotin and GBPNocotin shows that: GBNocotin is a new nicotianamine synthase gene, GBPNocotin contains a novel iron-regulated transcription factor.2. The expression of GBNocotin in different organs was studied using RT-PCR.The results showed that GBNocotin was expressed in leaves,stem, fiber, flowers except in immature seeds.3.The expression of GBNocotin under abiotic stress was detected by RT-PCR.The results showed the expression of GBNocotin is normal under PEG and GA treatment, but low under PEG、CuSO4and ABA treatment.4.The detection of PCR and restriction enzyme analysis shows that,the plant expression vector PBI121-E8-GBNocotin was constructed successfully,which contains fruit-specific promoter E8.5. The detection of PCR shows that,the plant expression vector pcxg-p-GBPNocotin was constructed successfully,which contains the GUS gene. By GUS staining,GBPNocotin promoter can drive GUS gene expression in different organs.
