Glyphosate is a broad-spectrum excellent herbicide for its high efficiency, low toxicity and residue. EPSPS is the target enzyme of glyphosate and it is the key enzyme in aromatic amino acid biosynthesis. Glyphosate interferes aromatic amino acid synthesis induce aromatic amino acid lacking. That causes weeds to die. This research was focused on the overexpression of EPSPS in Beta vulgaris Linn in order to make it grow normally after absorb glyphosate.Use AT cDNA as template and design primers which contained BamHI and SacI endonuclease site. Got complete AtEPSPs by PCR. Contruct plasmid vector pMD18-T-AtEPSPs and transform DH5a competent cells.Got Beta vulgaris aseptic seedings from8genotypes. Optimized agrobacterium-mediated transformation and established Beta vulgaris regeneration system. Inserted OsAPx8into Beta vulgaris by agrobacterium-mediated transformation. PCR showed31positive plants were got and positive rate was44.9%.
