Supramolecular chemistry as an important frontier discipline of chemistry developed from synthetic chemistry, information science, materials science, environmental science and life science. Supramolecular chemistry is a subject which surpasses the classic molecule and a highly complex chemical system created from the interaction force beyond the covalent bond.Molecular recognition is the research center of supramolecular chemistry; it is a specific process of combining host compounds with other gest molecules. The nitrogen-containing macrolides with nitrogen atoms and ester groups in the ring can be used as hosts in the study of molecular recognition. The nitrogen-containing macrolides have better stability than the chain compounds; also they are always characterized with specific combining properties because of the nitrogens and oxygens contained in the ring with good coordination ability.Ferrocene is characterized with unique properties in the aspects of structure, thermodynamics and medical science etc. It is a good choice to introduce the ferrocene as an unite into the macrocyclic compounds which can be expected to improve the overall properties such as energy and surface electrostatic potential of the molecule.This dissertation reviewed the significance and development in the supramolecular chemistry and medicine research of nitrogen-containing macrolides and ferrocenophanes. By analysis the structural features and properties of nitrogen-containing macrocycles, macrolides, nitrogen-containing macrolides and ferrocenophanes, we designed and synthesized ten novel nitrogen-containing macrolides and five ferrocenophanes. All the novel nitrogen-containing macrolides and ferrocenophanes were characterized by IR,’H-NMR, MS and Elemental Analysis, and the synthesis and molecular spectra of them were discussed. Some crystals were obtained to study and optimize the target compounds, also the crystal structures were analyzed and discussed. At the same time, the recognition properties and biological activities of some selected compounds were studied.
