Pyridine一2(1H)-thione and coumarin derivatives are potential bioactive molecules. Due to their signincant physiological activity they have been praised by the chemical workers of all ages.Pyridine-2(1H)-thiones contain N,S miscellaneous atomic,often show very well in antidepressant,anticancer.antiheart disease,and antimental.And coumarins have shown very well in anti-inflammatory,anticancer,and photo sensitive efiect.Both have a wide range of applications in the pharmaceutical,pesticide and other fields.Isonavones show a wide range of biological activities,such as anti-inflammatory diminish inflammation,anticancer,antiviral efiects.They as lealing compounds can condesated with triazole,2-aminobenzimidazole, and3-amino-5-hydroxypyrazole which contain two or three nitrogen atoms.Triazoopyrimidines, pyrimido[1,2-a] benzimidazoles, and pyrazolo[3,4-b]pyridines were obtained.Isoflavones as lealing compourlds can also condesated with cyanoacetamide and cyanoacetohydrazide which contain active methylene and nitrogen atoms.Pyridones and diazepines were obtained.First.we used cvanothioacetamide condesated with isoflavones in DMF in the present of sodium hydroxide.21novel pyridine-2(1H)-thiones were obtained and their struction were identitfied by the method of FT-IR,1H NMR,13C NMR.The crystal stru cture of5-phenyl-6-(2-hydroxy-4-isopropoxyphenyl)-3-cyanopyridine-2(1H)-thione was determined by single crystal X-ray diffraction.Second,we used propanedinitrile condesated with isoflavones in ethanol in the present of sodium methoxide.10novel4-benzyl coumarins were obtained and their struction were identitned by the method of FT-IR,1H NMR,13C NMR.Last,we used1,2,4-triazole condesated with isoflavones in DMSO in the present of n-BuOK.10novel1,2,4-triazolo[4,3-a]pyrimidin were obtained and their struetion were idelltitfied by the method of FT-IR,1H NMR,13C NMR.A novel strategy for the preparation of5,6-diary pyridine-2(1H)-thiones,4-benzyl coumarins and1,2,4-triazolo[4,3-a]pyrimidins by the one step condensation reaction with cyanothioacetamide,propanedinitrile and1,2,4-triazole was established.We provided a new way to produce pyridine-2(1H)-thione,4-benzyl coumarin,and1,2,4-triazolo[4,3-a]pyrimidins,and to speed up potential biological activity discovery of new drugs.
