Aspidistra belongs to the familiy of Liliaceae, Tillich recorded that there are about93species in the world in2008,which mainly distributed in Asia eastern tropical and subtropical areas. There are about67species in China at present, mainly distributed in the regions of the south of the Yangtze river basin in China,with Guangxi,Guizhou and Yunnan as her Natural Distribution Centre.Our study is focused on the species distributed in Guizhou,we have made systematic taxonomy research with them,including literatures acquisition work,wild investigation,samples collection,preparing of plant specimens, plant morphology study, cytotaxonomy research and DNA molecular identification. And the Flora of Guizhou were revised based on this study.With large amount of literatures acquisition work, wild investigation and samples collection,we revised Aspidistra sepcies in Flora of Guizhou,the new keys for identification of their botanical morphology were provided. There are15species in Guizhou at present,and replenished the13sepcies,such as A. fungilliformis,A. tonkinensis,A. oblanceifolia and A. sichuanensis,et al.,at the same time, one new species A. guizhouensis were proposed.The correlation between the karyotypes structure and morphological characteristics among seven species of the genus Aspidistra from Guizhou were investigated based on their chromosome numbers and external morphology, it can clarify the evolutionary origins of this species to provide a basis for classification and identification. Among all these seven species, one was found to have the chromosome number2n=36and the other six with2n=38. All the above species had bimodal karyotypes belonging to the Stebbins’ karyotype classification2C.The chromosome number and karyotype of A. pingtangensis, A. liboensis, A. chishuiensis and A. guizhouensis were reported for the first time. The results showed that in terms of nuclear structure and external morphology this genus has some relevance.It is the first time to use DNA molecular identification, To test the DNA molecular markers(matK、psbA-trnH、rbcL、ITS2、ITS、psbK-psbI and atpF-atpH) to identify medicinal plants in Aspidistra, the efficiency of PCR amplification and sequencing, intraspecific variation and interspecific divergence and identification efficiency were used to evaluate these loci. The results show that through analysis of21samples of12species of Aspidistra, rbcL、matK、psbA-trnH、psbK-psbI had high efficiency of PCR amplification and sequencing, the success identification rate of matK was the highest (85%) in single sequence. The discrimination ability of different sequences was shown higher using the sequence combinations, in which the success identification rate of matK+psbK-psbI was100%. Therefore,matK+psbK-psbI could be used to identify the medicinal plants in Aspidistra.
