[OBJECTIVE]The aim of this study is to optimize the extraction process of Baihuasandi,initially prove the relevance of dose and efficacy and establish the basis for preparing a better effective and convenient drug.[METHODS]In this Study, Three extracts were prepared for comparison of the antibacterial effect in vitro and pharmacological activity on CPID model rat.1.The antibacterial effect of the extracts of Baihuasandi in vitro were determined by test tube double dilution method. The MIC and MBC were used as evaluation.2.Injected bacteria into the left side of womb to make CPID model rat,48 female SD rats were selected and divided into 6 groups at random. The pathological change, engorgement of womb and the contents of IL-6 in the blood were observed to evaluate the influence of Baihuasandi exacts on model rats.3.Injected bacteria into the left side of uterui to make CPID model rat,60 female SD rats were selected and divided into 6 groups at random. The Pathological change, engorgement of womb and the contents of IL-6 in the blood were observed to evaluate the influence of Baihuasandi mingled decoction and than precipitation by alcohol on the model rat.[RESULTS]The Baihuasandi extracts all have antibacterial activity, the mingled decoction is as effective as the mingled decoction and than precipation by alcohol,and they are more effective than the mingled decoction after Zanthoxylum nitidum DC decocted with alcohol Staphylococcus against aureus and Bacillus coli.The three extracts have the same effect against streptococcus.Nitidine Chloride solution has no inhibitory effect against the three bacterium.CPID model rats were made successfully. All the treatment groups have therapeutic effects.The mingled group and the mingled and than precipation by alcohol group are better, and between them there was no significant difference. The further research shows us that low dose of the mingled and than precipation by alcohol group was effective. There was no significant difference between middle dose group and high dose group.[CONCLUSION]This study shows that the mingled decoction and the mingled and than precipation by alcohol group are better than the mingled decoction after Zanthoxylum nitidum DC decocted with alcohol.Nitidine Chloride is not the effective constituent of Zanthoxylum nitidum DC.The 1/2 of clinically effective dose of the mingled and than precipation by alcohol group can achieve the desired therapeutic effect, indicating that the normal clinical doses used before may be too large.In order to reduce drug side effects,improve economic efficiency and modifies drug types, the dose of medicine may be appropriately reduced.
