Diabetes has become the third chronic disease followed the disease of heart headblood-vessel and malignant tumor.and do seriously harm to human health.The pathogenesis oftypeⅡdiabetes is based on Insulin resistance, while obesity could enhance it. Understanding thegenerating process of lipocyte in vivo and identifying the molecular mechanism in differentiationof lipocyte can provide new ideas for the treatment to Diabetes.TRB3gene is the homologous gene with drosophila tribbles gene. Present research onTRB3is mainly focuses on insulin signaling pathways, NF-KB signaling pathways, BMPsignaling pathways, Lipid Metabolism and so on. It has been known that TRB3gene inhibits thedifferentiation of prelipocyte. ATF5, transcription activation factor5, is the member oftranscription activation factor family ATF/CREB, which is the effect element binding protein forCRE, containing basic amino acids zipper domain and DNA-binding domain.It was recentlyshowed that ATF5plays an important role on the differentiation, proliferation and survival ofmany tissues and cells. Effect of the interaction between ATF5and TRB3to the differentiationof prelipocyte was discussed in this paper aimed to provide new target points for treatment ofdiabetesExperimental methods:(1)Molecular cloning eukaryotic expression plasmid HA-TRB3、GFP-ATF5, constructingGFP-ATF5-△β–Zip and GFP-ATF5-β–Zip.(2)Indentifying the interaction between ATF5and TRB3by confocol、Luciferase、co-IP.(3)Obtaining cell lines able to transfer TRB3stably and Packaging adenovirus ATF5(4)Analysing effect of the interaction between ATF5and TRB3to the physiological indexesof mice withⅡdiabetes under high sugar stimulation condition, such as blood sugar.Results:(1)EukaryoticexpressionplasmidHA-TRB3,GFP-ATF5,GFP-ATF5-△β–Zip,GFP-ATF5-β–Zip were constructed Successfully.(2)The interaction between ATF5and TRB3was identified by confocol、Luciferase、co-IP experiment. Moreover, the β–Zip domain interaction between TRB3and ATF5was furtherdiscovered.(3) obtain th cell lines able to transfer TRB3stably and adenovirus ATF5was enclosed.(4)The interaction between ATF5and TRB3was proved to inhibit differentiation ofprelipocyte.Experimental conclusions:The interaction betweent ATF5and TRB3was proved firstly, besides, further resultsshowed that TRB3gene interacted with the β–Zip domain of ATF5.The results that differentiation of cell inhibited by the interaction betweent ATF5and TRB3were known in differentiation model research of ex-lipocyte...
