Objective: In recent years, the incidence of thyroid cancer increasedgradually, serious harm to human health, so people are increasinglyconcerned about the study of the pathogenesis of thyroid cancer. Thyroidcancer is mainly divided into papillary carcinoma, follicular carcinoma,medullary carcinoma, undifferentiated carcinoma, papillary carcinoma is themost common pathological type. Recent studies have shown that the incidenceof papillary thyroid cancer may be closely related to EB virus infection.EB virus is a double-stranded DNA virus, also known as human herpesvirus4. Adults around the world, about98%of the population is infectedwith EB virus, usually do not develop the disease, with or without clinicalmanifestations, and humanity is the sole host. Common diseases caused byEpstein-Barr virus infection: nasopharyngeal cancer, infectiousmononucleosis, Burkitt’s lymphoma, histiocytosis, T-cell lymphomas, suchas Hodgkin’s disease. In recent years, the research has focused on the role ofEB virus infection in liver cancer, lung cancer, breast cancer and other tumorless in papillary thyroid carcinoma. In addition, studies have shown that theincidence of papillary thyroid cancer may be activated RAS-RAF-MEK-ERKpathway, membrane expression after infection by the EB virus latent protein1(LMP1) may activate the pathway, and research has shown the pathwayexpression in benign thyroid lesions less. The ERK protein located in the pathof the final sites, its activation is considered a sign of the pathway activation,so this experiment was to investigate the EB virus infection activatedRAS-RAF-MEK-ERK pathway in papillary thyroid cancer role.Methods: Were collected in the Department of Pathology, the SecondHospital of Hebei Medical University from March2011to September2012,after the surgical removal of the thyroid disease archived paraffin blocks139 cases, including98cases of papillary carcinoma,41cases of thyroid adenoma.All thyroid cancer have not been preoperative radiotherapy and chemotherapytreatment. All thyroid tissue paraffin blocks were taken4μm thickness of afew pieces of serial sections, a slice routine HE staining for pathologicalmorphological observation, the other two for the Epstein-Barr virus detection,the use of in situ hybridization analysis of Epstein-Barr virus in papillarythyroid carcinoma and thyroid adenoma expression;2for ERK receptorimmunohistochemical staining and negative control, the SP method ERKreceptor in papillary thyroid carcinoma and thyroid adenoma expression andclinical data were statistically analyzed; SPSS statistical software usingchi-square test, nonparametric rank tests, linear correlation analysis and otherstatistical methods for data processing, test for P <0.05was statisticallysignificance.Results:1EBER-1expression in papillary thyroid carcinoma tissue was higherthan the thyroid adenoma, suggesting that Epstein-Barr virus infection may beclosely related to the occurrence of papillary thyroid carcinoma.2ERK expression in papillary thyroid carcinoma tissue was higher thanthe thyroid adenoma tissue, suggesting that the papillary carcinoma of thethyroid may occur with the Ras-Raf-Mek-Erk signal transduction pathway isactivated.3EBER-1in situ hybridization and ERK expression correlation studies:the the ERK expression data analysis of EB virus infection in63patients withpapillary thyroid cancer, EBER-1and ERK were positive in34cases,25were negative cases, both expression showed a positive correlation=5.866,P=0.0015).Conclusion:1EBER-1expression in papillary thyroid carcinoma tissue was higherthan normal thyroid tissue, suggesting that EB virus infection may be closelyrelated to the occurrence of papillary thyroid carcinoma.2ERK expression in papillary thyroid carcinoma tissue was higher than normal thyroid tissue, suggesting that the incidence of papillary thyroidcancer may be related to the Ras-Raf-MEK-ERK signal transduction pathwayis activated.3EBER-1expression and ERK expression results of the statisticalanalysis of the two was a positive correlation prompted EB virus infectionmay lead to the occurrence of papillary thyroid cancer by activating theRas-Raf-Mek-Erk signaling pathway.
