Objective Anti-tumor effort and function mechanism of curcumin,demethox ycurcumin and bisdemethox ycurcumin derived from Curcuma longa were compared to set up a foundation of development and utilizations on curcumin’s derivatives.Methods①Curcumin,demethox ycurcumin and bisdemethox ycurcumin were extracted with some methods,such as ethanol percolation extraction, petroleum ether degreasing, silica column chromatography isolation and acetone purification. Purity of curcumin and its derivatives was determinated by HPLC.②MTT reduction assay was used to observe the inhibiton of curcumin and its derivatives on HepG2; spectrophotometry was used to determine the influence on the activity of enzyme Caspase-3by curcumin and its derivatives. The genetic expression of HepG2Caspase-3was determined by RT-PCR; Expression of protein HepG2cells Caspase-3was determined by immunohistochemisty.③MTT reduction assay was used to observe the difference of proliferation the of HUVEC cells by curcumin and its derivatives; agarose curettage was used to determine the influence on the migration of HUVEC cells by curcumin and its derivatives; chick chorioallantoic membrane was used to observe the inhibiton of angiogenesis by curcumin and its derivatives.Results①The purity of curcumin,demethox ycurcumin and bisdemethox ycurcumin derived from Curcuma longa was more than90%by HPLC.②Three kinds of curcumins have suppressive function on HepG2cells and the more concentration demethox ycurcumin and bisdemethox ycurcumin were, the more suppressive function they had at the concentration ranging from6μmol·L-1to24μmol·L-1. Three kinds of curcumins strengthen the ability of enzyme Caspase-3in HepG2cells and high concentration’s bisdemethox ycurcumin was the best compared to curcumin,demethox ycurcumin. Immunohistochemisty indicated that Three kinds of curcumins strengthen the expression of Caspase-3protein in HepG2cells and high concentration’s bisdemethox ycurcumin was the best compared to curcumin,demethox ycurcumin which was in line with mRNA expression of Caspase-3.③Three kinds of curcumins significantly exert suppressive effects on proliferation and migration of HUVEC cells. At high concentration, the performance of inhibition on migration of HUVEC cells from three kinds of curcumins was that curcumin was strongest and demethox ycurcumin was stronger than bisdemethox ycurcumin. Chick chorioallantoic membrane indicated that three kinds of curcumins exerted inhibitions on angiogenesis to some extent. Curcumin exerted significant inhibition on angiogenesis at middle and high doses. Bisdemethox ycurcumin exerted more significant inhibition on angiogenesis than curcumin.Conclusions①we had extracted high purity of curcumin and its natural derivatives(demethox ycurcumin and bisdemethox ycurcumin).②three kinds of curcumins exerted inhibitions on HepG2cells and stimulated the expression of Caspase-3. Bisdemethox ycurcumin had strongest inhibition compared to curcumin and demethox ycurcumin that may be one of mechanism stopping HepG2cells growing in vitro.③Curcumin and its derivatives,especially bisdemethox ycurcumin, exerted inhibition on angiogenesis.
