Objective Observe the effects of treatment in repairing skeletal muscle after Acute Contusion process with myosin heavy chain; MHC, collagen-Ⅰ and collagen-Ⅲ with Huoxuezhitong cream and massage.Methods Using homemade muscle injury device to caused66Japanese white rabbits bilateral lower extremities after the gastrocnemius contusion, and then the animals were randomly divided into five groups as the Huoxuezhitong ointments high, medium, low group, massage and control groups and trauma control group. Each group was then divided into four groups according to the observation point of time, the number of samples in each group of three. Another six the normal rabbit will be undamaged for doing normal control. Animal injury therapy treatment and control group approach same as before. Damage after3,10,20and27days, respectively, with3%pentobarbital sodium ear vein injection (40mg/kg) rabbits anesthesia remove bilateral gastrocnemius muscles of the damaged area, and stored at-20℃refrigerator. Using semi-quantitative reverse transcription-polymerase chain reaction to(semi-quantitative RT-PCR) analysis the MHC-Type Ⅱb, Ⅰ and type Ⅲ collagen mRNA expression.Results①Animals in each group after Acute Contusion skeletal muscle II b MHC class I and III collagen expression increased with the upward trend in the repair process and maintained at a relatively constant level will drop late to repair and maintain the final constant level.②The paste group on Huoxuezhitong rat type Ⅱb MHC protein expression levels in skeletal muscle repair process increases and the peak was significantly higher than the low-dose group (P<0.05), but lower than that of the high group (P<0.05), and a massage roughly the same group, but eventually a constant value of massage the group than Huoxuezhitong cream group.③5rats group in type I collagen protein levels peaked10days after injury, but the paste Huoxuezhitong rats the protein peak was significantly lower than the trauma group and low dose group (P<0.05), significantly higher in the high-dose group, and the massage group showed no significant difference, the massage group constant value is smaller than the trauma group (P<0.05) and remaining constant value was no significant difference.④Rats Ⅲ collagen expression levels were reached its peak in the first10days after injury, dropped to a certain level of basic maintenance at a higher than normal level. The Huoxuezhitong paste dose group rat type Ⅲ collagen protein peak level was significantly lower than the low-dose group and trauma group (P<0.05) higher than the high-dose group, compared to the massage group had no significant difference (P<0.05); final constant value was no significant difference between groups.Conclusion Chinese medicine and massage of the muscle contusion repair process the Huoxuezhitong class can effectively promote the type Ⅱb MHC protein, prevents Ⅰ excessive increase in the expression of type Ⅲ collagen, thus contributing to the repair of damaged tissue.
