Objective:Testicular torsion is one of the common urological emergencies requesting appropriate treatments on time.Otherwise,it would result in injury on testis.Quercetin,one of the flavonoids,has been recognized as possessing extensive clinical effects,such as anti-inflammatory,antioxidant and antitumoral activities.This study was to investigate the effects of quercetin on injured testis by establishing a model of tcsticular torsion/detorsion in rats.Methods:A total of54male Sprague-Dawley rats were randomly divided into A,B and C group,respectively.Testicular torsion was created by rotating the left testis in a clockwise direction.The tissue image was evaluated with light microscopy by implication of Makler score method,and levels of both SOD and MDA were valued, and reverse transcription PCR(RT-PCR) was applied to detect the relative genetic expression of bcl-2and bax.In addition, we utilized the flow cytometer to compute the apoptosis ratio of germ cells,as well as levels of sexual hormone in blood serum in each groups respectively.Results:Compared with group B and C,repectively,tissue dmages was detected obviously and Makler scores were significantly decreaed in group A(P<0.01).Results in group B,compared with those of group A,showed an more increased testicular structures and reasonable levels of both serum sex hormone and SOD, higher expression of bcl-2, lower levels of MDA,gene expression of bax, as well as apoptosis ratio, respectively(P<0.05).In addition, animals in group B showed an improved histological appearance and Makler score than that of group A. Our data also indicated a significant reduction in the capacity of MDA,expression of bax gene and apoptosis ratio,an improvement of expression of bax gene and testicular structures,level of both Makler score and SOD, and more reasonable sex hormone levels in group C, compared with those of group B, and the differences were statistically remarkble (P<0.05).Conclusions:Testicular histological structures and functions will inevitablely suffer an reduction in different degrees during the ischaemia-reperfusion period.Furthermore, quercetin possesses the capacity of eliminating reactive oxygen species and inactivating lipid peroxidation with the result of keeping the balance between oxidant system and anti-system;it can also serve as repressing the tansductional process of apoptotic signal,as well as maintaining the stabilization among FSH, LH and T, which work together to prevent the testis from testicular torsion/detorsion-induced injury.
